Background/Aims: CTLA-4 polymorphisms are associated with susceptibility to various cancers, but the results are often conflicting. Hence, we performed a comprehensive meta-analysis to quantitatively investigate the association between CTLA-4 polymorphisms (rs231775, rs4553808，rs5742909, rs3087243 or rs733618) and cancer risk. Methods: Data were collected from PubMed and Web of Science. A total of 67 case-control studies were selected for quantitative analysis. Stata (Version 12) software was used to calculate the odds ratio (OR) and 95% confidence interval (CI) to evaluate the strength of the associations. Subgroup meta-analysis was conducted based on ethnicity and cancer type. Heterogeneity tests, sensitivity analysis, and publication bias assessments were also performed. Results: rs231775, rs4553808 and rs5742909 but not rs3087243 and rs733618 were significantly related to cancer risk. In analyses stratified by ethnicity, both rs231775 and rs4553808 were significant susceptibility polymorphisms in an Asian population but not in a Caucasian population. Moreover, there were stronger associations between the rs231775 polymorphism and increased risk of bone, breast, liver, head and neck and pancreatic cancers. Additionally, rs4553808 was associated with significantly increased susceptibility to breast cancer and head and neck cancer. Conclusion: rs231775, rs4553808 and rs5742909 may be used as predictive genetic biomarkers for cancer predisposition. Combined detection of CTLA-4 SNPs could be a useful tool for prediction of cancer susceptibility in clinical practice.

The treatment of advanced or refractory non-small cell lung cancer has been historically difficult owing to the lack of studies on effective systemic cure. The progress in lung cancer treatment has plateaued, necessitating new options for additional benefits. Immune checkpoint proteins are co-inhibitory factors that can diminish the antigen-specific immune responses by attenuating the regulatory role of cytotoxic T-lymphocyte-associated protein 4, programmed cell death-1, lymphocyte-activation gene 3, and T-cell immunoglobulin mucin-3. The therapeutic strategies targeting immune checkpoints mainly focus on the monoclonal antibody of these regulatory factors, which may facilitate clinical decision making. An enhanced understanding of the drug-resistance mechanisms and the therapeutic efficacy regulation will provide opportunities to improve the clinical outcomes of non-small cell lung cancer patients. Preclinical and clinical trials on these key immune-regulatory agents, which has heralded a new era in immuno-oncology in non-small cell lung cancer treatment, are currently in development. Abstract. Cytotoxic T-lymphocyte associated antigen-4 (CTLA-4) polymorphisms have been examined for associations with cervical cancer in various countries. The results, however, are inconclusive. The present study aimed to explore whether CTLA-4 +49 A/G, -318 C/T and CT60 G/A polymorphisms confer susceptibility to cervical cancer. A meta-analysis was performed with 7,794 subjects included in 15 case-control studies that were published up to January 1, 2013. The results from the meta-analysis indicated that there were no significant associations between the risk of cervical cancer and the three studied polymorphisms [+49 A/G: Odds ratio (OR), 0.94 and 95% confidence interval (CI), 0.82-1.07 for GG+AG vs. AA; -318 C/T: OR, 1.33 and 95% CI, 0.82-2.16 for TT+TC vs. CC; and CT60: OR, 0.98 and 95% CI, 0.72-1.33 for AA+AG vs. GG]. Stratified analyses by ethnicity for the +49 A/G and -318 C/T polymorphisms suggested that Asian populations had a decreased risk of cervical cancer for the +49 A/G polymorphism (OR, 0.75 and 95% CI, 0.58-0.97 for GG+AG vs. AA), but an increased risk for the -318 C/T polymorphism (OR, 2.02 and 95% CI, 1.36-3.00 for TC vs. CC). In summary, the current meta-analysis showed that the +49 A/G and -318 C/T polymorphisms in CTLA-4 constitute risk factors for cervical cancer.
