CSN5/Jab1 Is Involved in Ligand-Dependent Degradation of Estrogen Receptor α by the Proteasome

Abstract: Here, we show that estrogen receptor ␣ (ER␣) coimmunoprecipitates with CSN5/Jab1, a subunit of the COP9 signalosome (CSN), and that overexpression of CSN5/Jab1 causes an increase in ligand-induced ER␣ degradation. Inhibition of either the kinase activity associated with the CSN complex by curcumin or of nuclear export by leptomycin B (LMB) impaired estradiol-induced ER␣ degradation by the proteasome. Degradation of ER␣ induced by the pure antagonist ICI 182,780 (ICI) was blocked by curcumin but not by LMB, ind… Show more

“…While the mechanisms involved in ER commitment to proteasomal degradation and the role of the latter process in ER-mediated gene transactivation have been under extensive investigation (Nawaz & O'Malley 2004, Calligé & Richard-Foy 2005, Dennis & O'Malley 2005, comparatively little attention has been paid to a possible compartmentalization of the system(s) responsible for ER elimination. The issue was addressed in the present study by investigating the impact of LMB on ER distribution, downregulation, and transactivation activity in MCF-7 breast carcinoma cells.…”

“…Jab1 in the nucleus functions as transcription coactivator and may interact and modulate the activity of steroid receptors as demonstrated previously. 33,34 Our results indeed demonstrated a marginal correlation between Jab1 and estrogen receptor or progesterone receptor ( Table 2). The functional interaction between steroid receptors and Jab1 received much attention recently.…”

“…Chauchereau et al 34 firstly reported that Jab1 interacted with progesterone receptor and its coactivator SRC-1 which resulted in stabilization of the progesterone receptor/SRC-1 complex. Subsequently, Callige et al 33 showed that Jab1 was involved in ligand-dependent degradation of estrogen receptor by the proteasome. These data suggested that Jab1 might modulate the transcriptional activity of steroid receptors.…”

“…It was later found to be a regulator of various factors including p27 Kip1 , cyclin E, Smad family, and SCF ubiquitin ligases (17)(18)(19)(20)(21)(22). In particular, the CSN complex targets p53 for its phosphorylation and facilitates its degradation via the ubiquitin system (23).…”
