Cs + Induces the kdp Operon of Escherichia coli by Lowering the Intracellular K + Concentration

Abstract: Cs؉ was found to induce expression of the kdpFABC operon, encoding a high-affinity K ؉ uptake system of Escherichia coli. Quantitative expression analyses at the transcriptional and translational levels reveal that CsCl causes much higher induction of kdpFABC than does NaCl. A decrease of the intracellular K ؉ concentration is found in cells exposed to CsCl. The results indicate that kdpFABC expression is induced when the intracellular K ؉ concentration is lowered. Moreover, the results imply that the signal t… Show more

“…It has been proposed that the kdp expression is regulated by the internal K ϩ concentration and medium osmolarity, 5,21) but no direct evidence for this has been provided until now. Our present study provided the direct evidence to show that kdp expression is regulated by the internal K ϩ concentration.…”

“…This technique reveals only relative induction levels by using a radioactively labeled probe that is specific for the mRNA of interest. The protocol used for Northern analysis was described previously (21). Briefly, 20-g samples of RNA were fractionated by electrophoresis on 1.2% (wt/vol) agarose gels containing 1.1% (vol/vol) formaldehyde in MOPS (4-morpholinepropanesulfonic acid) buffer.…”

“…The KdpD/KdpE system controls expression of the kdpFABCDE operon, which encodes the high-affinity K ϩ transporter, as well as KdpD and KdpE (18,77). The expression of this operon is inducible by both NaCl and CsCl via different mechanisms (33). The entire kdpFABCDE operon was deleted in the KdpD/KdpE mutants.…”
