Crystallization and preliminary X-ray studies of recombinant amylosucrase fromNeisseria polysaccharea

Abstract: Recombinant amylosucrase from Neisseria polysaccharea was crystallized by the vapour-diffusion procedure in the presence of polyethylene glycol 6000. The crystals belong to the orthorhombic space group P2 1 2 1 2, with unit-cell parameters a = 95.7, b = 117.2, c = 62.1 A Ê , and diffract to 1.6 A Ê resolution. A p-chloromercuribenzene sulfonate (pcmbs) derivative has been identi®ed and a selenomethionine-substituted protein has been produced and crystallized.

“…In CD2, the corresponding dipeptide NV is replaced with 2214 FI, which can also be found in the amylosucrase sequence. Structural data obtained for this enzyme (52) suggest that the Phe residue could be engaged in the specificity towards the fructo-furanosyl ring of sucrose (53). Moreover, the Ile residue at position 2215 is also found in GTF-A from L. reuteri (59) and is strongly conserved in the ␣-amylase family.…”

“…This led to a new classification of the glucansucrase enzymes in family 70 of the glycoside hydrolases on the CAZy website (http://afmb.cnrs-mrs.fr/ϳpedro/CAZY/db.html). The only exception is amylosucrase from Neisseria polysaccharea, which belongs to family 13 since its structure is not circularly permutated and is closely related to the ␣-amylase structure with a much lower molecular size than that of the glucansucrase enzymes (40,52).…”

“…For family GH13 enzymes, high-resolution three-dimensional structures are available, often in complex with substrates, inhibitors, or products (207). The three-dimensional structures of amylosucrase (also in complex with sucrose, oligosaccharides, and with a covalently bound glucopyranosyl moiety) resemble those of other proteins of family GH13 (80,115,181,183). Sequence alignments, mutagenesis studies, and three-dimensional structure analyses within family GH13 have resulted in identification of the amino acid residues involved in catalysis (204).…”

“…However, Neisseria polysaccharea amylosucrase (NpAS) is the only AS for which several structures, alone or in complex with sucrose substrate or products, are available to date (17)(18)(19)(20). The three-dimensional structure of NpAS is organized in five domains, namely A, B, C, N, and BЈ.…”
