Crystal structure of a phosphatase-resistant mutant of sporulation response regulator Spo0F from Bacillus subtilis

Abstract: The structural analysis reveals that the overall topology and metal-binding coordination at the active site are similar to those of the bacterial chemotaxis response regulator CheY. Structural differences between Spo0F and CheY in the vicinity of the active site provide an insight into how similar molecular scaffolds can be adapted to perform different biological roles by the alteration of only a few amino acid residues. These differences may contribute to the observed stability of the phosphorylated species o… Show more

“…One of the water molecules was hydrogen bonded to the main chain carbonyl oxygen of Gln-55. The coordination of the metal to the third acidic residue (Glu-9) and the absence of direct coordination to the main chain carbonyl oxygen of Gln-55 differs from what has been reported for all metal-bound receiver domains (32)(33)(34)(35)(36).…”

“…YneI is most probably a single-domain response regulator. The sequence of this 120-residue protein is very similar to that of CheY and Spo0F, for which the three-dimensional structures are known (22,40). YneJ is a predicted 160-residue integral membrane protein without clear similarity to any protein sequence available in the databases.…”

“…This section focuses on the crystallographic results, and NMR studies are discussed in a different section. Structural analysis has been carried out on the metalfree form (17) and on the metal-bound forms (18,19). Spo0F, like all other response regulators, has an ␣/␤ fold with a central ␤-sheet and five helices surrounding it (Fig.…”
