Crystal structure and molecular dynamics of human POLDIP2, a multifaceted adaptor protein in metabolism and genome stability

Abstract: Polymerase δ-interacting protein 2 (POLDIP2, PDIP38) is a multifaceted, ‘moonlighting’ protein, involved in binding protein partners from many different cellular processes, including mitochondrial metabolism, DNA replication and repair, and reactive oxygen species generation. POLDIP2 is found in multiple cellular compartments, potentially shuttled depending on its role. How POLDIP2 binds to and coordinates many different proteins is currently unknown. Towards this goal, we present the crystal structure of the … Show more

“…DNA polymerase delta interacting protein 2 (POLDIP2) was initially characterized as an accessory factor for several error-prone nuclear DNA polymerases. It primarily localizes to the mitochondrial matrix, however, and critical contacts for activity with DNA polymerase are removed by mitochondrial targeting sequence cleavage, suggesting that it has a distinct function in the mitochondrion (10). POLDIP2 has also been identified as an interactor and possible regulator of CLPX (11,12), but the mechanism by which POLDIP2 might regulate CLPX activity has not been determined.…”
