Cryptosporangium eucalypti sp. nov., an actinomycete isolated from Eucalyptus camaldulensis roots

Abstract: A novel actinomycete, designated strain EURKPP3H10T, was isolated from surface-sterilized roots of Eucalyptus camaldulensis Dehnh., collected from Kamphaengphet Silvicultural Research Station, Kamphaengphet province, Thailand. The taxonomic position of strain EURKPP3H10T was studied using a polyphasic approach. Phylogenetic evaluation based on 16S rRNA gene sequence analysis showed that strain EURKPP3H10T belongs to the genus Cryptosporangium, with the highest sequence similarity to Cryptosporangium cibodasens… Show more

“…Among the seven genera, Cryptosporangium spp. has also been isolated from plant litter [102,103] or as endophytes of plants [104]. Due to the close phylogenetic relationships among known species and between many of our samples and those known species, the final taxonomic placements of most of our isolates, especially the yellow-highlighted ones, such as S3-21 and S3-31, in Figure 6, await further investigation using either multilocus sequence typing or whole-genome sequencing.…”

“…A 0.1 ml aliquot of the various dilutions was spread on humic acid–vitamin agar [9], final pH 7.0, supplemented with nalidixic acid (25 µg ml −1 ) and nystatin (50 µg ml −1 ). After incubation at 28 °C for 14 days, colonies of the strains were isolated and purified on glucose–yeast extract agar (GYEA) [10]. Mycelia and spores of the strains were preserved in sterile glycerol solution (20 % v/v) at −20 °C.…”

“…A 0.1 ml aliquot of the various dilutions was spread on humic acid-vitamin agar [5], final pH 7.0, supplemented with nalidixic acid (25 µg ml −1 ) and nystatin (50 µg ml −1 ). After the plates were incubated at 28 °C for 14 days, the single colony of strain RB6PN24 T was transferred and purified on glucose-yeast extract agar (GYEA) [6]. The strain was subsequently maintained and preserved on GYEA slant at 30 °C.…”
