Crosstalk between keratinocytes and immune cells in inflammatory skin diseases

Abstract: Cutaneous homeostasis is maintained by dynamic cellular communications between different cell types in the skin through interactions with various mediators, including cytokines, chemokines and antimicrobial peptides/proteins (AMPs). Keratinocytes, as the major cell type of the epidermis, not only form a passive physical barrier, but also actively participate in the pathogenesis of many, if not all, inflammatory skin diseases. Keratinocytes highly interact with immune cells to shape, amplify or regulate inflamm… Show more

“…A growing number of recent studies have demonstrated that keratinocytes in psoriasis interact with immune cells to form a feed-forward inflammatory loop. They act as a trigger and amplifier of the inflammatory response by recruiting and activating immune cells, thereby triggering local inflammation, which plays important roles in the initiation, maintenance, and relapse phases of psoriasis ( 64 ).…”

“…In AD lesions, cytokines released by inflammatory cells exacerbate inflammation [ 14 , 34 ]. Hence, we measured the gene expression levels of IL-4, IL-5, and IL-13 as Th2 cytokines, IFN-γ and IL-12 A as Th1 cytokines, and IL-1β as a pro-inflammatory cytokine in ear tissue of AD-induced mice using RT- q PCR.…”

“…When AD progresses to the chronic phase, Th1 response is also increased with the secretion of interferon (IFN)-γ [ 13 ]. Immune cells recruited into skin lesions secrete inflammatory mediators that activate keratinocytes and exacerbate inflammatory responses [ 14 ]. Accordingly, regulating the inflammatory responses of keratinocytes and immune cells is the main therapeutic approach for AD.…”

“…Specific stimuli induce keratinocytes to produce a range of inflammatory molecules for the homing of immune cells in AD skin lesions. TARC and MDC are expressed strongly by keratinocytes and are involved in the recruitment of 2 lymphocytes [55]. In patients with AD, the levels of TARC and MDC, as well as their receptor C-C chemokine receptor type 4 (CCR4), are higher than those in normal individuals.…”
