Cross-Talks between Natural Killer Cells and Distinct Subsets of Dendritic Cells

Abstract: In recent years, the essential role of bi-directional cross-talk between natural killer (NK) and dendritic cells (DC) during immune responses has been clearly elucidated. In particular, this cross-talk results in the development of an efficient innate response, through DC-mediated NK cell activation, and a potent adaptive immune response, through NK-mediate DC editing and maturation. Recently, some novel human DC subsets have been identified: migratory DCs in afferent lymph and draining lymph nodes; CLEC9A+/BD… Show more

“…Such "DC-editing" by NK cells occurs both in peripheral tissues and in secondary lymphoid organs (36). This is well in accord with our observations of Ad5/3-D24-tras-treated mice showing redistribution of NK cells toward tumors and draining lymph nodes, coinciding with accumulation of activated DCs and reduction in immature DCs.…”

“…Interestingly, NK cell depletion by anti-asialo antibody completely blunted this effect (P ¼ 0.037, as compared with Ad5/3-D24-tras), indicating a central role for NK cells in editing/stimulating DCs, as has been previously suggested (36). A similar trend was observed with regard to CD80-single-positive DCs.…”

“…Accordingly, enhanced killing of K562 and lymphoma cells by NK effectors could be demonstrated upon stimulation of lymphocytes with tumor cellloaded IFN-DC. Although the cross-talk between NK cells and mature DC has been largely described (19,20), it should be pointed out that, in contrast to classical monocyte-derived DC, IFN-DC were shown not to need any additional factors to behave like mature DC (7,10) and spontaneously produce chemokines known to promote NK cell recruitment (21). Moreover, differentiation of monocytes into IFN-DC in the presence of NK cells was previously shown to favor a more mature DC phenotype (22).…”
