Cross-breed comparisons identified a critical 591-kb region for bovine carcass weight QTL (CW-2) on chromosome 6 and the Ile-442-Met substitution in NCAPG as a positional candidate

Abstract: Background: Growth-related traits have been mapped on bovine chromosome 6 (BTA 6) in various bovine breed populations. We previously mapped a significant quantitative trait locus (QTL) for carcass and body weight (CW-2) between 38 and 55 cM on BTA 6 using a Japanese Black half-sib family. Additional QTL mapping studies detected four QTL for body or carcass weight that overlapped with CW-2 in Japanese Black and Japanese Brown half-sib families. To map the region in greater detail, we applied cross-breed compari… Show more

“…The LD block in Japanese Black encompassed a 591‐kb interval including FAM184B , DCAF16 , NCAPG and LCORL and non‐synonymous amino acid substitution Ile442Met (pI442M) in NCAPG was found as a candidate causative variation (Setoguchi et al . 2009). The NCAPG pI442M allele was shared in the different cattle breeds used in these studies.…”

“…In our analysis, one of the Japanese Black sires segregating the carcass weight QTL was homozygous between 39.2 and 61.0 Mb on BTA6 (UMD3.1), therefore the region was excluded (Setoguchi et al . 2009). …”

“…These results of CWT and REA were consistent with data obtained in previous study in the Kagoshima prefecture population [7], and thus confirmed that the NCAPGc. 1326T>G SNP is a causative variation for bovine CWT [2].…”

“…A previous study showed that the c.1326T>G single nucleotide polymorphism (SNP) in the NCAPG (non-SMC condensin I complex, subunit G) gene, which leads to the amino acid change of Ile442 to Met442 in the encoded protein, was identified as a candidate causative variation for a bovine carcass weight (CWT) quantitative trait locus (QTL) on bovine chromosome 6 [2]. The NCAPG c.1326T>G variation is present in various bovine breeds and has linkage or association with birth weight [3] [4], and withers height and body weight at adolescence [4]- [6].…”
