Critical role of nitric oxide in the modulation of prepulse inhibition in Swiss mice

Abstract: This study provides the first demonstration that an increase in NO can improve the PPI response and block the amphetamine-disruptive effects on the PPI response. Our data are consistent with recent clinical results. However, these effects do not appear to be related to an increase in cGMP levels, and further investigation is thus required.

“…Indeed, it has been speculated that NO exerts a tonic inhibitory effect on dopamine transporters, which could correct a reduced activity of dopamine in the prefrontal cortex and, through feedback loops, fix dopaminergic hyperactivity in both accumbens and striatum (Pycock et al, 1980). Interestingly, Issy et al (2014) have recently shown in rats that pretreatment with SNP attenuated schizophrenia-like changes in PPI (prepulse inhibition) induced by amphetamine, a dopaminergic agonist (Issy et al, 2014).…”

“…Bujas-Bobanovic et al 4 described that SNP was able to inhibit the schizophrenia-like behavior and brain c-fos expression in rats induced by PCP, an NMDA receptor antagonist. Also, Issy et al 20 reported that SNP attenuated the schizophrenia-like changes in prepulse inhibition in rats induced by amphetamine, a dopaminergic agonist.…”

“…Several adverse effects such as nausea, vomiting, diarrhea, agitation, headache, insomnia, somnolence, anxiety, and sexual dysfunction are among the most frequently reported adverse effects of SSRIs. 6,7 However, bleeding is a rare condition related to SSRI treatment. There are some case reports describing young or adult patients with various bleeding problems (eg, ecchymosis, purpura, epistaxis, gastrointestinal, or vaginal bleeding) related to SSRI treatment.…”

“…2Y4 Bujas-Bobanovic et al 5 reported that a nitric oxide (NO) donor, sodium nitroprusside (SNP), produced rapid improvement in NMDA receptor blockadeYinduced ''psychosis'' in rats. Issy et al 6 showed that SNP increases the prepulse inhibition baseline in a dose-dependent manner and also blocks the prepulse inhibition disruption induced in the amphetamine model of psychosis. It has also been reported that pluripotent stem cells reprogrammed from fibroblasts of patients with schizophrenia and then differentiated into neurons showed decreased postsynaptic density protein 95 levels and glutamate receptor expression.…”
