Critical Appraisal of the MTT Assay in the Presence of Rottlerin and Uncouplers

Abstract: Rottlerin is a natural product isolated from Mallotus philippinensis. This polyphenolic compound, originally described as a selective inhibitor of PKCδ, can inhibit many other PKC-unrelated kinases and has a number of biological actions, including mitochondrial uncoupling effects. We recently found that Rottlerin inhibits the transcription factor nuclear factor κB in different cell types, causing downregulation of cyclin D1 and growth arrest. The present study was carried out to clarify the surprising lack of … Show more

“…The MTT assay is based on the colorimetric conversion of yellow 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide to the purple formazan product (22) and was employed to assess cell viability in the present study. PC-12 cells were seeded in 96-well plates with a density of 0.5 ϫ 10 5 cells/well.…”

“…Drugs and chemical agents can also significantly modify MTT-specific activity if these treatments increase or decrease mitochondrial respiration or cause metabolic uncoupling. Drug-induced respiratory uncoupling will produce a large false high, whereas agents that decrease metabolic respiration and electron transport will produce a false low in MTTspecific activity as compared to vehicle-treated controls containing identical cell numbers (3,10).…”
