Critical and Multiple Roles for the CD3ε Intracytoplasmic Tail in Double Negative to Double Positive Thymocyte Differentiation

Abstract: The preTCR is associated with signal-transducing CD3␥, ␦, , and polypeptides. It is generally agreed that CD3 chains play redundant roles in the receptor-mediated signal transduction. In the present study, we show that the intracytoplasmic (IC) domain of CD3 is essential for early thymocyte maturation. We demonstrate that the IC domain-deleted CD3 fails to restore the double negative (DN) to double positive (DP) thymocyte development in CD3-deficient mice. Additional experiments show that the membrane proximal… Show more

“…of the suboptimal deletion of the Nck2 flox alleles in DN thymocytes. Notably, a partial block at the DN3 stage is shared by other deletional models of direct and indirect binding partners of Nck; namely, WASP (32), Vav (33), and the CD3ε.PRS, although, in this case, in some (13,22) but not all (12,14) models investigated.…”

“…The Nck adapters have been implicated in the regulation of the surface levels of the TCR (11,22,23). The levels of TCR expression are finely regulated at the DP stage (24), as both positive and negative selection depend on the strength of TCR signaling (25).…”

“…Bone marrow reconstitution with CD3ε PRS mutants in CD3ε-deficient mice, Nck overexpression in primary T cells, Nck knockout mice, and PRS knockin (KI) mice have all been used to study the role of the PRS and Nck in T cell development and mature T cell activation (14)(15)(16)(17)(18)(19)(20). Some experiments have suggested that the PRS is important for thymic maturation, but not for mature T cell activation in vitro (15), although experiments with bone marrow chimeras indicate that the PRS is important to activate mature T cells by weak, but not by strong, agonists in vitro (19).…”
