CRH Promotes the Neurogenic Activity of Neural Stem Cells in the Adult Hippocampus

Abstract: Local cues in the adult neurogenic niches dynamically regulate homeostasis in neural stem cells, whereas their identity and associated molecular mechanisms remain poorly understood. Here, we show that corticotropin-releasing hormone (CRH), the major mediator of mammalian stress response and a key neuromodulator in the adult brain, is necessary for hippocampal neural stem cell (hiNSC) activity under physiological conditions. In particular, we demonstrate functionality of the CRH/CRH receptor (CRHR) system in mo… Show more

“…The alternative to the single factors presented above is suggested by several protocols that propose the use of chemical compound "cocktails". There are attempts to use mixes containing factors stimulating neuronal differentiation, such as forskolin, insulin, hydrocortisone, and valproic acid in the media [25,44], ongoing both with the maintenance of the cells' high proliferation rate. In this study, we analyzed the impact of one of them that was not previously used in ASCs' differentiation, commercially available supplement N21.…”

“…Data represent the mean ± SEM. *P ≤ .05 quantification of SOX2-and doublecortin-positive cells, series of every sixth brain section were stained and positive cells were counted throughout the rostro-caudal extent of the granule cell layer(Koutmani et al, 2019).…”

“…In the newborn rat, the PVN and CA1 of the hippocampus contain GR mRNA expression ( Pryce, 2008 ). The CRH hippocampal system regulates neurogenesis in the hippocampus which is involved in spatial learning and memory ( Koutmani et al, 2019 ). Elevated levels of glucocorticoids have also been shown to impair working and reference memory ( Stylianakis et al, 2018 ).…”
