CRF-R1 activation in the anterior-dorsal BNST induces maternal neglect in lactating rats via an HPA axis-independent central mechanism

Abstract: HighlightsCRF-R1 activation in the adBNST reduces arched back nursing and nursing.adBNST CRF-R2 activation increases arched back nursing but impairs overall nursing.Inhibiting adBNST CRF-R1 prevents the stress-induced decline in arched back nursing.CRF-R activation in the adBNST stimulates the HPA axis in a stress-dependent manner.CRF-R-induced HPA axis activation is not responsible for reduced maternal care.

“…Activation of CRF-R postpartum impairs the expression of maternal behavior thereby leading to maternal neglect (D'Anna and Gammie, 2009;Gammie et al, 2004;Klampfl et al, 2014Klampfl et al, , 2016Klampfl et al, 2013;Pedersen et al, 1991). Hence, hypoactivation of CRF-R during this sensitive period is an indispensable prerequisite for the appearance of appropriate maternal behavior.…”

“…Maternal care was monitored on lactation day (LD) 1 before and after substance infusion (non-stress condition) as well as on LD 5 before and after substance infusion, which was combined with a psychosocial stressor (i.e. the maternal defense test; stress condition Bayerl et al, 2014;Klampfl et al, 2014Klampfl et al, , 2016Klampfl et al, 2013)). Observations were conducted for 10 s every 2 nd min in 30 min blocks according to an established protocol (Bosch and Neumann, 2008).…”

“…Importantly, the brain CRF system also modulates various social behaviors, like mother–infant interaction (Gammie et al 2004; Klampfl et al 2013, 2014, 2016) or pair bond formation in male prairie voles. In the latter, central activation of the CRF system facilitates pair bond formation, even in the absence of mating (DeVries et al 2002).…”
