Creating Defined Gaseous Environments to Study the Effects of Hypoxia on C. elegans

Abstract: Oxygen is essential for all metazoans to survive, with one known exception 1 . Decreased O 2 availability (hypoxia) can arise during states of disease, normal development or changes in environmental conditions [2][3][4][5] . Understanding the cellular signaling pathways that are involved in the response to hypoxia could provide new insight into treatment strategies for diverse human pathologies, from stroke to cancer. This goal has been impeded, at least in part, by technical difficulties associated with contr… Show more

“…H 2 S Exposure-C. elegans were exposed to H 2 S in atmospheric chambers perfused with H 2 S continuously diluted into room air, as described (15). Concentrated tanks of compressed H 2 S gas (5,000 ppm balanced with N 2 ) were purchased from Airgas.…”

“…In contrast, C. elegans does not have a circulatory system, relying instead on diffusion for O 2 delivery to cells. This allows for precise experimental control of both genotype and cellular environment (Shen and Powell-Coffman, 2003; Fawcett et al, 2012). Because it is an attractive model for hypoxia research we have built a framework of hypoxia responses as a function of O 2 tension using C. elegans , drawing connections with other systems when possible.…”

“…This protocol presents two ways to perform the experiments, one using a lab-made apparatus (using Tupperware-type containers and gas input 11 ) and other using a commercial hypoxic chamber 4,6 . Anoxia can be achieved by other means that consume the oxygen, as described elsewhere 13,17 …”
