Cre recombinase-mediated restoration of nigrostriatal dopamine in dopamine-deficient mice reverses hypophagia and bradykinesia

Abstract: A line of dopamine-deficient (DD) mice was generated to allow selective restoration of normal dopamine signaling to specific brain regions. These DD floxed stop (DDfs) mice have a nonfunctional Tyrosine hydroxylase (Th) gene because of insertion of a Neo R gene flanked by lox P sites targeted to the first intron of the Th gene. DDfs mice have trace brain dopamine content, severe hypoactivity, and aphagia, and they die without intervention. However, they can be maintained by daily treatment with L-3,4-dihydroxy… Show more

“…Adult Foxa1/2 cKO mice also show reduced preference for a sweetened solution, which may be indicative of a reduced drive for rewarding stimuli. DA role in feeding is well documented as DA-deficient mice are hypoactive and hypophagic (38,39), pharmacological blockade of DA signaling in the striatum correlates with impaired feeding (63), and restoration of DA signaling in DA-deficient mice promotes feeding behavior (64). To distinguish direct roles of FOXA1/2 in regulating feeding behavior from motor activity, we have conducted motor function and activity assays.…”

“…Dopamine-deficient mice (Th fs/fs , Dbh Th/ϩ ) carrying two conditionally inactive Th alleles, were generated as described (18). Dopamine-deficient mice are indistinguishable from littermates at birth and maintain normal growth when given daily L-dihydroxyphenylalanine (L-dopa) treatment (30 mg/kg) that restores locomotion for Ϸ8 h (18). Animal protocols were approved by the Institutional Animal Care and Use Committee.…”

“…KOR (Oprk1) gene deletion on C57BL/6 background was generated as previously described (23), and homozygous (KOR KO) and WT (KOR WT) littermate controls were generated by heterozygote crosses. Dopamine-deficient mice (Th fs/fs , Dbh Th/ϩ ) carrying two conditionally inactive Th alleles, were generated as described (18). Dopamine-deficient mice are indistinguishable from littermates at birth and maintain normal growth when given daily L-dihydroxyphenylalanine (L-dopa) treatment (30 mg/kg) that restores locomotion for Ϸ8 h (18).…”

“…Licking data were processed and grouped into 2-h bins by using the MatLab software (Mathworks). Meal number and meal size were analyzed as described (50), and the average lick volume was 1.07 l per lick.…”
