Crabp2 Promotes Metastasis of Lung Cancer Cells via HuR and Integrin β1/FAK/ERK Signaling

Abstract: Increased Crabp2 levels have been found in various types of cancer, and are associated with poor patients’ survival. Although Crabp2 is found to be overexpressed in lung cancer, its role in metastasis of lung cancer is unclear. In this study, Crabp2 was overexpressed in high-metastatic C10F4 than low-metastatic lung cancer cells. Analysis of clinical samples revealed that high CRABP2 levels were correlated with lymph node metastases, poor overall survival, and increased recurrence. Knockdown of Crabp2 decrease… Show more

“…Previous studies have linked known cancer drivers to RBP dysregulation. For example, the oncogene crabp2 interacts with the RBP HuR to promote metastasis of lung cancer cells by regulating integrin β1/FAK/ERK signaling (Wu et al, 2019). Transforming growth factor-β (TGF-β) induces the expression of RNA-binding motif protein 38 (RBM38) in breast cancer, which promotes epithelial-to-mesenchymal transition by regulating the zonula occludens-1 transcript (Wu et al, 2017).…”

“…Yu S et al report that CRABP2 promotes the invasion of pancreatic cancer cells through stabilizing the interleukin 8 expression [34]. Wu J et al demonstrate that CRABP2 enhances lung cancer metastasis by HuR and integrin β1/FAK/ERK signaling [35]. These results highlight the oncogenic role of CRABP2 in the progression of cancers.…”

“…In addition, activation of ITGA7 is correlated with metastasis, recurrence, and reduced survival in colorectal cancer patients . Although the function of ITGA7 is still unclear in NSCLC, there has been study illustrating that the activation of integrin beta 1 (ITGB1), with whom ITGA7 binds and forms a heterodimeric adhesion receptor, and its downstream signaling molecules FAK and ERK promote resistance to apoptosis, enhance cell migration, and invasion of lung cancer cells . In addition, the FAK/AKT singling pathway, which is activated by ITGA7 is responsible for lung cancer cell migration and invasion as well .…”
