CpG Oligodeoxynucleotides and Interleukin-12 Improve the Efficacy ofMycobacterium bovisBCG Vaccination in Mice Challenged withM. tuberculosis

Abstract: Mycobacterium bovis bacillus Calmette-Guérin (BCG) is the only vaccine approved for prevention of tuberculosis. It has been postulated that serial passage of BCG over the years may have resulted in attenuation of its effectiveness. Because interleukin-12 (IL-12) and oligodeoxynucleotides (ODN) containing cytidine phosphate guanosine (CpG) motifs have been shown to enhance Th1 responses in vivo, they were chosen as adjuvants to increase the effectiveness of BCG vaccination. In this report, mice were vaccinated … Show more

“…TLR9 was subsequently recognized as a receptor for CpG DNA (35). Internalization of CpG DNA and intracellular trafficking play key roles in signaling (24,29). Much research has focused upon the roles of adaptor proteins and downstream kinases that lead to activation of AP-1 and NF-B transcription factors.…”

“…3e). Previous studies (29) suggested that IL-12 treatment synergizes with CpG DNA to promote cellular responses. To test for IL-12 synergy, monocytes were incubated for 60 min with 50 ng/ml IL-12, followed by addition of murine or human CpG DNA.…”

“…It is likely that host cytokine cascade may have the most pronounced and long-lasting effects on the T cells' responses. Recently, Freidag et al (66) reported that IL-12 improved the efficacy of BCG vaccination in mice challenged with M. tuberculosis. Kremer et al (67) showed that intradermal injection with DNA encoding IL-18 led to increased in vitro PPD-dependent IFN-␥ production in mice infected with BCG.…”

“…However, several studies have shown that intranasal delivery, with CpG oligodeoxynucleotides as adjuvants, results in strong systemic and mucosal immune responses to co-administered antigens, including hepatitis B surface antigen [14] and influenza virus [33]. Intranasal administration of CpG and interleukin-12 also improves the efficacy of BCG vaccination in mice challenged with M. tuberculosis [34].…”
