[(Cp-R)M(CO)3] (M= Re or 99mTc) Conjugates for Theranostic Receptor Targeting

Abstract: Cyclopentadienyl complexes of 99mTc became accessible via a retro Diels-Alder synthetic approach of dimerized cyclopentadiene derivatives. So far, this approach was limited to derivatives comprising a carboxylic acid group, directly conjugated to the Cp-ring, leading to complexes [(C5H5COOH)99mTc(CO)3] and [(C5H5CONH-R)99mTc(CO)3], respectively. The introduction of an -NCO group via Curtius rearrangement and subsequent in situ reactions with alcohols or amines gave [(C5H5NHCO-OR)2] and [(C5H5NHCO-NHR)2]. To in… Show more

“…The metal chosen for creating the model radiopharmaceutical complexes presented in this paper is rhenium, which is ideally suited due to its similarities to technetium. 9 In radiopharmacy technetium is used as a diagnostic agent utilizing the 99m Tc isotope which decays to release gamma radiation. 99m Tc is routinely used in thyroid, 10 heart, 11 kidney, 12 brain 13 and bone 14 imaging; moreover the isotope is investigated for selective cancer imaging.…”

“…Re compounds [15,[21][22][23]. Due to the small size of octahedral complexes based on the the fac-[M(CO) 3 ] + (M = Re and Tc) core considerably to the octahedral or square-pyramidal complexes of the corresponding metals in higher oxidation states, they are considered less likely to impact important characteristics of bio-molecules to which they are conjugated to [14,24].…”

“…29,34,50 The “matched-pair theory,” in which the Tc and Re isotopes are utilised for tandem therapeutic and diagnostic (theranostic) applications, has also become an attractive concept in the field of nuclear medicine. 52–54 This offers the possibility of developing 99m Tc agents for patient pre-selection and follow-up by diagnostic imaging and, thereafter, treatment with the corresponding 186/188 Re analogues.…”
