The rates of plankton community production and respiration were determined from in vitro changes in dissolved inorganic carbon and dissolved oxygen and the incorporation of NaH14C03 at a coastal site in East Antarctica between 16 December 1993 and 12 February 1994. The breakout of seasonal fast ice was associated with a succession of dominant phytoplankton from Cryptomonas to Phaeocystis to a d~atom assemblage. Gross production reached 33 mm01 C m-3 d-l and I4C incorporation peaked at 24 mm01 C m-3 d-' on 23 January 1994, at the time of the chlorophyll a maximum (22 mg chl a m-3). Dark community respiration reached its maximum (13 mm01 C m-3 d-' ) 4 d later. Photosynthetic rates calculated from I4C incorporation were s~gnificantly lower (17 to 59%) than rates of gross production. The derivation of plankton processes from changes in both dissolved oxygen and dissolved inorganic carbon allowed the direct measurement of photosynthetic and respiratory quotients. A linear regression of all data gave a photosynthetic quotient of 1.33 + 0.23 and a respiratory quotient of 0.88 * 0.14. Concurrent determination$ of bacterial, heterotrophic dinoflagellate, nanoflagellate and ciliate respiration could account for 15 to 58% of measured dark community respiration. This study has improved the sparse data set of plankton respiration measurements, confirmed that heterotrophic respiration is a significant process in the carbon flux of coastal Antarctic waters and achieved a first apportionment of community respiration to the major microbial groups in this region

Con el propósito de estimar la productividad primaria del fitoplancton mediante el método de oxígeno disuelto (AO,) con diferentes tiempos de incubación y compararlo con la estimada mediante el método de carbono catorce ('"C), se realizaron simultáneamente experimentos de productividad primaria con ambos métodos, los días 12 de noviembre de 1993, 3 y 16 de febrero de 1994, y 24 de mayo de 1994. La productividad primaria bruta (PPB) estimada por el método de AO, fue similar a los valores derivados por el método de '"C, con a = 0.05. Además, para conocer la variabilidad de la concentración de clorofila y abundancia bacteriana, se recolectó agua de mar cada 30 min durante el periodo de muestreo. Se concluye que, debido a su abundancia, es importante considerar las bacterias en estudios encaminados a estimar la productividad primaria en cuerpos costeros, ya que afectan el resultado del método de AOZ, por la utilización del oxígeno por parte de las bacterias cuando hay suficiente sustrato dentro de las botellas. Los valores de la PPB estimada con el método de AO, pueden ser recomendadas como una alternativa viable al método de 14C para situaciones donde no es conveniente o posible el uso de isótopos radioactivos, siempre y cuando se realicen los experimentos con réplicas y por lo menos con tres tiempos finales de incubación.

Anaerobic anoxygenic phototrophic bacteria can be found in the suboxic waters of shallow stratified coastal systems, and may play important roles in the total primary production of subtropical stratified coastal lagoons. We investigated the spatiotemporal variability of light CO(2) fixation and net oxygen production in the stratified Conceição Lagoon (Brazil) in summer and fall of 2007, as well as the contribution of bacteriochlorophyll a (BChl a)-containing bacteria to photosynthetically driven electron transfer. Both chlorophyll a (Chl a) and BChl a varied in space, while only BChl a varied in time (three-fold increase from summer to fall). In summer, net oxygen production and light CO(2) fixation were correlated, with both having higher rates with higher Chl a concentrations in the enclosed region of the lagoon. In fall, CO(2) fixation was decoupled from oxygen production. Denaturing gradient gel electrophoresis revealed that bacterial communities of oxic site 12 and suboxic site 33 formed one cluster, different from other oxic samples within the lagoon. In addition, BChl a/Chl a ratios at these sites were high, 40% and 45%, respectively. Light acted as the main factor controlling the BChl a concentration and CO(2) fixation rates. High turbidity within the enclosed area of the lagoon explained high BChl a and decoupling between CO(2) fixation and oxygen production in oxygenated waters. Contribution of purple sulfur bacteria to total bacterial density in suboxic waters was 1.2%, and their biomass contributed to a much higher percentage (12.2%) due to their large biovolume. Our results indicate a significant contribution of anaerobic anoxygenic bacteria to the primary production of the "dead zone" of Conceição Lagoon.
