Preliminary safety estimate of cosmetic anti-age creams with chokeberry extract, using in vivo bioengineering techniques

Potential of anti-age cosmetic line, day and night creams were evaluated using noninvasive measurements of the appropriate skin biophysical parameters in a 24-h in vivo study under occlusion. Thirtyone healthy female volunteers were involved in the study. Electrical capacitance (EC), transepidermal water loss (TEWL), erythema index (EI) were evaluated. The preliminary safety profile of the investigated creams with natural origin ingredients based on obtained results can be considered satisfactory. The stated samples did not cause the change in the measured biophysical parameters-TEWL and EI, while they increased EC i.e. stratum corneum hydration. These results suggest that chokeberry extract and other natural ingredients could exert beneficial effects and has potential on aged skin thereby slowing down the appearance of wrinkles and their existing alleviation, and they could be considered preferable in the cosmetic products intended for the treatment of aged skin.

In Situ Gel Formation in Microporated Skin for Enhanced Topical Delivery of Niacinamide

Although used widely in cosmetic formulations, topical delivery of niacinamide (LogP = −0.35) is unfavorable by conventional means. Poly(lactide-co-glycolide) (PLGA) formulations, can undergo a sol-gel transition triggered by solvent exchange, entrapping molecules and sustaining their release. The current study aims to exploit the ability of PLGA to gel in situ and enhance the topical delivery of niacinamide in microporated skin. In vitro drug permeation studies were performed using vertical Franz diffusion cells. Microporation was performed using Dr. PenTM Ultima A6, where pre-treatment with a 1 mm needle-length for 10 s and a 0.5 mm needle-length for 5 s, both at 13,000 insertions/min were compared. The effect of different grades of PLGA, EXPANSORB® DLG 50-2A (“low” molecular weight), and EXPANSORB® DLG 50-8A (“high” molecular weight) on topical delivery was also determined. Formulations containing PLGA resulted in successful gelation in situ on application over microporated skin. A significantly higher amount of drug was found in the skin with the 0.5 mm treatment for 5 s (892 ± 36 µg/cm2) than with 1 mm for 10 s (167 ± 16 µg/cm2). Hence, the different grades of PLGA were evaluated with 0.5 mm, 5 s treatment, and a significantly larger amount was seen in skin with the higher rather than the lower molecular weight polymer (172 ± 53 µg/cm2).

BackgroundHyperpigmentation is a skin disorder characterized by elevated production of melanin. Current treatment approaches mainly rely on the application of skin lightening chemicals, most of which have safety issues. Efficacy of delivery of the active ingredients to the target organ has also been a challenge. Transdermal based drug delivery platform has been shown to improve drug bioavailability, avoiding the hepatic first pass metabolism, decrease gastrointestinal side effects, and eventually enhance patient compliance.ResultsThis article explores the utilization of micellar transdermal delivery technology to improve skin penetration and efficacy of arbutin, a hyperpigmentation agent. The suppression efficacy of cellular melanin production versus cell viability of four active ingredients commonly used in skin lightening products, namely allantoin, arbutin, glycolic acid, and hyaluronic acid were first compared. Arbutin was selected for the micellar delivery studies base on its comparatively low cytotoxicity and better performance in reducing melanin production. Micellar Arbutin cream was formulated using Urah® proprietary micellar technology and was assessed for its cellular melanin suppression efficacy and skin penetration capacity.ConclusionThe results show that micellar arbutin cream improved both the delivery and cellular melanin suppression, suggesting that micellar transdermal delivery may have potential application in addressing hyperpigmentation skin disorders.Graphical abstractTransdermal delivery of arbutin with micelles for melanin production suppression.Electronic supplementary materialThe online version of this article (doi:10.1186/s13104-016-2047-x) contains supplementary material, which is available to authorized users.
