Corydalis edulis Maxim. Promotes Insulin Secretion via the Activation of Protein Kinase Cs (PKCs) in Mice and Pancreatic β Cells

Abstract: Corydalis edulis Maxim., a widely grown plant in China, had been proposed for the treatment for type 2 diabetes mellitus. In this study, we found that C. edulis extract (CE) is protective against diabetes in mice. The treatment of hyperglycemic and hyperlipidemic apolipoprotein E (ApoE)−/− mice with a high dose of CE reduced serum glucose by 28.84% and serum total cholesterol by 17.34% and increased insulin release. We also found that CE significantly enhanced insulin secretion in a glucose-independent manner … Show more

“…The HIT-T15 cells were a suitable clone for screening plant extracts for sulfonylurea-like activity via insulin secretion and sulfonylurea receptor 1 (SUR1)-binding studies [81]. In another study, HIT-T15 cells treated with whole plant extracts of Corydalis edulis displayed enhanced glucose-independent insulin secretion [136].…”

“…Glucagon-like peptide-1 (GLP-1), an endogenous glucoselowering hormone, also stimulates insulin secretion through the PLC/PKC-dependent pathway in isolated islets (40). The extract from Corydalis edulis Maxim., a widely grown plant in China, was found to enhance insulin secretion by the activation of PKC in pancreatic β cells (41). The results provide evidence for the protective role of PTF against PA-induced impairment of GSIS involving GPR40 signaling in INS-1 cells.…”

“…HIT-T15 pancreatic islet cells were provided by Prof. Guan Y. F. (Peking University, Beijing, People’s Republic of China). The procedure for the assay of insulin secretion was the same as previously described, and each experiment was performed three times . Gliclazide (100 μM) was utilized as a positive control.…”
