Corticotropin releasing activity of the new CRF is potentiated several times by vasopressin

Abstract: Initially the hypothalamic factor responsible for the release of corticotropin (CRF), was thought to be a simple peptide. More recent work has led to the conclusion that CRF is a multifactorial complex. In 1979 we proposed that vasopressin, much disputed as a CRF candidate, was a major constituent of the complex, interacting with a potentiating the CRF activity of the other component(s). The recent characterization of a 41 residue ovine hypothalamic peptide capable of releasing adrenocorticotropic hormone (ACT… Show more

“…In addition, AVP released from the SON may induce ACTH release from the pituitary [59] . CRH and AVP stimulate ACTH secretion from pituitary corticotrophs by interacting with the CRH type 1 (CRHR1) and Avpr1b or V1b receptors, respectively [62] .…”

“…Some CRH neurons in the PVN co-express and release AVP. When released together into the hypophyseal-portal system, AVP strongly potentiates the ACTH-releasing activity of corticotrophs [59,60] .…”

“…After the discovery of CRH (Vale et al 1981) it was rapidly established that AVP potently synergizes with CRH to stimulate pituitary ACTH secretion: when CRH and AVP are co-administered, hormone output is well above the combined effects of the two neuropeptides in both humans (von Bardeleben and Holsboer 1985) and rodents (Gillies et al 1982). AVP is mainly synthesized in the magnocellular neurons of the hypothalamic PVN and the supraoptic nucleus.…”

“…CRF is synthesized and secreted in the hypothalamic paraventricular nucleus (PVN) in response to stress, and subsequently stimulates adrenocorticotropic hormone (ACTH), which is encoded by the pro-opiomelanocortin (POMC) gene in pituitary corticotrophs (Gillies et al, 1982;Mouri et l., 1993). ACTH stimulates the release of glucocorticoids from the adrenal glands, and these glucocorticoids play a variety of roles in the target tissues (Whitnall, 1993).…”
