Corticosteroid-induced kidney dysmorphogenesis is associated with deregulated expression of known cystogenic molecules, as well as indian hedgehog

Abstract: An intact genome is essential for kidney growth and differentiation, but less is known about whether, and how, an altered fetal milieu modifies these processes. Maternal low-protein diets perturb growth of the metanephros, the precursor of the mature kidney. Fetal corticosteroid overexposure may, in part, mediate this, because such diets downregulate placental 11beta-hydroxysteroid dehydrogenase-2, which degrades maternal corticosteroids. We report that glucocorticoid and mineralocorticoid receptors are expres… Show more

“…Pictures of whole stained kidneys were taken at low magnification under a dissecting microscope and the average area of individual cysts (defined as dilated tubules .0.01 mm 2 in cross-sectional area) was determined using ImageJ particle analysis (http://rsbweb.nih.gov.ij/). 26 Immunohistochemistry was performed for CD206 (MCA2235T, AdB Serotec, Oxford, UK) with appropriate secondary antibodies conjugated to horseradish peroxidase and detected by 3,39-diaminobenzidine with the color development performed for the same duration of time for each sample. Negative controls consisted of omission of primary antibodies or substitution with preimmune serum.…”

“…Based on previous studies (7,12,13), a relationship between metanephric size expansion and ureteric tip count, a more direct measurement of renal development, was expected. However, as can be noted in Figure 1a, surface area expansion did not correlate with ureteric tip formation (R 2 = 0.005, P = 0.77) and was therefore not used as a marker in our studies.…”

“…Finally, a study examining the effects of corticosteroid overexposure on metanephric development found that adding hydrocortisone to a metanephros organ culture unexpectedly caused renal cysts as well as upregulated Ihh transcripts. 16 Addition of Hh inhibitor, cyclopamine, reduced hydrocortisoneinduced cysts without affecting organ growth, implicating increased Hh signaling in this mechanism of cystogenesis.…”
