A growing body of psychiatric research has emerged, focusing on the role of endocannabinoid system in psychiatric disorders. For example, the endocannabinoid system, via cannabinoid CB (CB1 and CB2) receptors, is able to control the function of many receptors, such as N-methyl-d-aspartate (NMDA) receptors connected strictly with psychosis or other schizophrenia-associated symptoms. The aim of the present research was to investigate the impact of the CB1 receptor ligands on the symptoms typical for schizophrenia. We provoked psychosis-like effects in mice by an acute administration of NMDA receptor antagonist, MK-801 (0.1–0.6 mg/kg). An acute administration of MK-801 induced psychotic symptoms, manifested in the increase in locomotor activity (hyperactivity), measured in actimeters, as well as the memory impairment, assessed in the passive avoidance task. We revealed that an acute injection of CB1 receptor agonist, oleamide (5–20 mg/kg), had no influence on the short- and long-term memory-related disturbances, as well as on the hyperlocomotion in mice, provoking by an acute MK-801. In turn, an amnestic effects or hyperactivity induced by an acute MK-801 was attenuated by an acute administration of AM 251 (0.25–3 mg/kg), a CB1 receptor antagonist. The present findings confirm that endocannabinoid system is able to modify a variety of schizophrenia-like responses, including the cognitive disturbances and hyperlocomotion in mice. Antipsychotic-like effects induced by CB1 receptor antagonist, obtained in our research, confirm the potential effect of CB1 receptor blockade and could have important therapeutic implications on clinical settings, in the future.

Cognitive and motor disturbances are serious consequences of tremor induced by motor disorders. Despite a lack of effective clinical treatment, some potential therapeutic agents have been used to alleviate the cognitive symptoms in the animal models of tremor. In the current study, the effects of WIN55, 212-2 (WIN), a cannabinoid receptor (CBR) agonist, on harmaline-induced motor and cognitive impairments were studied. Adult rats were treated with WIN (0.5mg/kg; i.p.) 15min before harmaline administration (10mg/kg; ip) after which exploratory and anxiety related behaviors, and cognitive function were assessed using open-field behavior and shuttle box tests. Rats that received harmaline only exhibited a markedly reduced number of central square entries when compared to harmaline vehicle-treated controls, whereas those treated with WIN and harmaline showed a significant increase in central square entries, compared to harmaline only treated. The passive avoidance memory impairments observed in harmaline treated rats, was reversed somewhat by administration of WIN. The neuroprotective and anxiolytic effects of WIN demonstrated in the current study can be offered cannabinoid receptor (CBR) agonism as a potential neuroprotective agent in the treatment of patients with tremor that manifest mental dysfunctions.

Background: The Hot Plate test is a thermal hyperalgesia model to assess the analgesic activity of many compounds. Traditional analysis compares reaction times with controls, reports only analgesic activity without considering escape reflex. Escape reflex to pain sensation also provides an indirect measure of analgesic activity. However, it requires repeated exposure of rats to hot plate. We explored the learning activities and escape behaviour of rats by calculating the change from baseline of reaction times.Methods: Male Wister strain albino rats, weighing between 150-200mg were used for assessment of hot-plate induced hyperalgesia. Author analysed the analgesic activity of aspirin (100mg/Kg) and compared with animals receiving saline. Aspirin and saline were administered 1hr orally before the commencement of the experiment. The reaction time was recorded at every 30 min up to 6hrs.Results: With traditional analysis, we can observe that the onset of action, peak action and duration of analgesic activity of aspirin was at 0.5, 2.0 and 5.0 hours respectively. Peak inhibition of reaction time was seen at 2 hours with a magnitude of 474%. When we applied a statistical procedure, absolute change from baseline statistics, we found that animals treated with aspirin also exhibited escape reflex after fading of drug response and animals treated with saline demonstrated early (0.5hr) escape behaviour from the hot plate and continued throughout the rest of the experiment.Conclusions: Change from baseline as an outcome illustrates escape behaviour of rats in hot plate method and it should be employed along with percentage inhibition during assessment of analgesic activity.
