Increased cured leaf yield is an important breeding target for flue‐cured tobacco (Nicotiana tabacum L.), but genetic improvement for this trait has slowed in the last thirty years. Yield is positively correlated with days to flowering and leaf number in this species. Use of novel allelic diversity influencing these traits might contribute to further yield improvement. In this study, an introgressed quantitative trait locus (QTL) affecting flowering time and leaf number, designated as Many Leaves (Ml), was backcrossed into three genetic backgrounds of flue‐cured tobacco. Field evaluation of nearly isogenic MlMl lines and Mlml hybrids indicated that the QTL acted in an additive to partially dominant fashion to delay flowering and increase leaf number. This resulted in significant increases in yield and cash return per hectare. As the zygosity of Ml increased, corresponding decreases in percentage total alkaloids and increases in percentage reducing sugars were observed, however. Incorporation of Ml into flue‐cured tobacco breeding programs may be useful for increasing cured leaf yields in certain environments. Concurrent modification of the genetic potential to accumulate alkaloids would be also important under current industry requirements, however.

Yellowing is a key stage in the curing of flue-cured tobacco (Nicotiana tobacum L.) as much of the chemical transformation occurs during this period. This study examined the effect of different yellowing degrees on the value of flue-cured tobacco leaves at the farm level for both processing and manufacturing. The study was conducted in the counties of Chuxiong, Dali, and Yuxi in Yunnan, China over two years. Yellowing treatments have been designed to have either a mild or a regular yellowing degree. Yield, value, appearance, suction property, smoking characteristics, and physical resistance to further processing were investigated to evaluate the effect of degree of yellowing on the industrial utilization of flue-cured tobacco leaves. The regular yellowing degree enhanced yield, value, and appearance compared to the mild yellowing degree, regardless of cultivar or location; however, physical resistance to further processing and the suction property of the mild yellowing degree treatment were better than with the regular yellowing degree regardless of cultivar or location. Furthermore, although the regular yellowing degree recorded higher smoking characteristic scores than the mild yellowing degree immediately after flue-curing, the scores of mild yellowing degree leaves could be further augmented by increasing intensity in the re-drying stage. The smoking characteristic score in the regular yellowing degree can only be increased by low intensity re-drying, and significantly decreased by mild and high intensity re-drying. Therefore, in terms of industrial utilization, mild yellowing is the better choice for flue-curing tobacco. This study also suggested that the current regular yellowing stage in Yunnan should be shortened to meet the demands of the traditional tobacco industry.

The tobacco growing area is of great importance in the quality control of cigarette, because the fragrance of tobacco leaves would be divergent for different climates planting environments. Currently, most of discrimination processes are manually operated, which are time-consuming and inevitably limited by the subjective evaluation. In this paper, an automatic growing area discrimination method is presented based on tobacco nearinfrared (NIR) spectrum using support vector machine (SVM). The Savitzky-Golay smoothing method and principle component analysis are used for tobacco NIR spectra preprocessing. A SVM model is established to investigate the characteristics of growing areas. The developed SVM classifier produces the best prediction accuracy of 80.3% in testing subset with 14 principle components as the inputs. It is 6% and 2% higher than that of artificial neuron network and Mahalanobia distance model respectively, which were developed for comparison. It demonstrates the effectiveness and robustness of SVM for growing area discrimination. The prediction ability for each growing region is further analyzed by the measurements derived from confusion matrix, such as true positive rate, true negative rate, positive predictive value and F1 score. The SVM setting is also discussed with respect to prediction accuracy of validation.
