Correlation between loss of PTEN expression and PKB/AKT phosphorylation in hepatocellular carcinoma

Abstract: The clinical significance of phosphatase and tensin homolog deleted on chromosome ten (PTEN) protein expression and the correlation between the expression of PTEN and phosphorylation of protein kinase B (PKB/AKT) in human hepatocellular carcinoma (HCC) were investigated. The expression of PTEN and phospho-AKT was detected by SP immunohistochemical technique and Western blotting in 35 cases of HCC, 15 cases of liver cirrhosis and 8 cases of normal tissues. The correlation between the expression of PTEN and PKB/… Show more

“…We therefore wanted to scrutinize the effect of miR-17-5p and its inhibitor on its direct (PTEN and E2F1) as well as indirect targets (Wip1 and MAPK14) within sorafenib resistant EGFR/IL-6 signaling cascade based on the evidences provided by various studies. PTEN, a gatekeeper tumor suppressor gene has been reported to exert its tumor suppressing function via negatively regulating PI3K/Akt cell survival signaling pathway by dephosphorylating the D3 position of PIP3 47 , 48 . Activated PI3K/Akt pathway has been observed to play a significant role in promoting acquired resistance to sorafenib in HCC 30 , 49 .…”

“…Activated PI3K/Akt pathway has been observed to play a significant role in promoting acquired resistance to sorafenib in HCC 30 , 49 . Furthermore, PTEN has also been shown to be significantly reduced in normal liver tissues compare to HCC liver tissues 48 . Huang et al ., reported significantly shorter overall survival in HCC patients with decreased expression of PTEN 50 .…”
