BackgroundThe prevalence, symptom course, and shedding in persons infected with the 4 most common human coronaviruses (HCoV)‐229E, HKU1, NL63, and OC43 are poorly described.ObjectivesWe estimate their prevalence and associated symptoms among college students identified via a social network study design.Patients/MethodsWe collected 1‐3 samples (n = 250 specimens) from 176 participants between October 2012 and January 17, 2013: participants with acute respiratory infection (ARI; cough and body aches or chills or fever/feverishness) and their social contacts. Virus was detected using RT‐PCR.Results30.4% (76/250) of specimens tested positive for any virus tested, and 4.8% (12/250) were positive for 2 or more viruses. Human coronaviruses (HCoVs [22.0%; 55/250]), rhinovirus (7.6%; 19/250), and influenza A (6.4%; 16/250) were most prevalent. Symptoms changed significantly over time among ARI participants with HCoV: the prevalence of cough and chills decreased over 6 days (P = .04, and P = .01, respectively), while runny nose increased over the same period (P = .02). HCoV‐NL63 was the most frequent virus detected 6 days following symptom onset (8.9%), followed by rhinovirus (6.7%).ConclusionsDuring a 3‐month period covering a single season, HCoVs were common, even among social contacts without respiratory symptoms; specific symptoms may change over the course of HCoV‐associated illness and were similar to symptoms from influenza and rhinovirus.

Structural investigation of ACE2 dependent disassembly of the trimeric SARS-CoV-2 Spike glycoprotein

The human membrane protein Angiotensin-converting enzyme 2 (hACE2) acts as the main receptor for host cells invasion of the new coronavirus SARS-CoV-2. The viral surface glycoprotein Spike binds to hACE2, which triggers virus entry into cells. As of today, the role of hACE2 for virus fusion is not well understood. Blocking the transition of Spike from its prefusion to post-fusion state might be a strategy to prevent or treat COVID-19. Here we report a single particle cryo-electron microscopy analysis of SARS-CoV-2 trimeric Spike in presence of the human ACE2 ectodomain. The binding of purified hACE2 ectodomain to Spike induces the disassembly of the trimeric form of Spike and a structural rearrangement of its S1 domain to form a stable, monomeric complex with hACE2. This observed hACE2 dependent dissociation of the Spike trimer suggests a mechanism for the therapeutic role of recombinant soluble hACE2 for treatment of COVID-19.

Aromatik halkasında bir veya daha fazla hidroksil grubu içeren bileşikler fenolik madde olarak adlandırılmaktadır. Fenolik maddeler bitkilerin meyve, sebze, yaprak, tohum, çiçek, gövde ve dallarında bulunmaktadır. Fenolik maddeler antioksidan özelliklerine bağlı olarak antikanserojen, antimutajenik, antimikrobiyal aktivite göstermektedir ve insan sağlığı ile yakından ilişkilidir. Zeytin, zeytin yaprağı ve zeytinyağı fenolik içerikleri ile önemli bileşiklerdir. Oleuropein, zeytin meyvesinin olgunlaşmadan önceki döneminde bol miktarda bulunan, meyve olgunlaştıkça miktarı azalan ve meyveye acı tat veren bir maddedir. Kuersetin; lahanalar, soğan, kırmızı üzüm ve çayda bol miktarda bulunan polifenolik bir flavonoiddir. Diyette en fazla bulunan flavonoid olan kuersetin diyet suplemanları, yiyecek ve içeceklere acı tat veren bir maddedir. Çalışmalar, oleuropein ve kuersetinin organizmadaki zararlı maddelerin hücre yapısını bozarak sağlık için olumlu etkiye sahip olduğunu göstermektedir. COVID-19 koronavirüs ailesinin son tanımlanan türlerinden biridir. Tedavi ve korunma yöntemi bulunmayan hastalığa karşı insanlar çeşitli çözümler denemektedir. Bu bağlamda başvurulan yollardan önemli bir tanesi de fenolik bileşiklerdir. Bu çalışmada, dünya genelinde birçok insanın hayatını kaybetmesine neden olan COVID-19’ a karşı oleuropeinin ve kuersetinin etkinliğinin belirlenmesi amaçlanmaktadır.
