Since the severe acute respiratory syndrome (SARS) outbreak in 2003, human coronaviruses (hCoVs) have been identified as causative agents of severe acute respiratory tract infections. Two more hCoV outbreaks have since occurred, the most recent being SARS-CoV-2, the causative agent of coronavirus disease 2019 (COVID-19). The clinical presentation of SARS and MERS is remarkably similar to COVID-19, with hyperinflammation causing a severe form of the disease in some patients. Previous studies show that the expression of the SARS-CoV E protein is associated with the hyperinflammatory response that could culminate in acute respiratory distress syndrome (ARDS), a potentially fatal complication. This immunemediated damage is largely caused by a cytokine storm, which is induced by significantly elevated levels of inflammatory cytokines interleukin (IL)-1β and IL-6, which are partly mediated by the expression of the SARS-CoV E protein. The interaction between the SARS-CoV E protein and the host protein, syntenin, as well as the viroporin function of SARS-CoV E, are linked to this cytokine dysregulation. This review aims to compare the clinical presentation of virulent hCoVs with a specific focus on the cause of the immunopathology. The review also proposes that inhibition of IL-1β and IL-6 in severe cases can improve patient outcome.

Zusammenfassung. Hintergrund: Schutzmaßnahmen zur Eindämmung der COVID-19-Pandemie waren mit Isolationsmaßnahmen in der stationären Altenpflege verbunden. Aufgrund der bekannten negativen Auswirkungen von sozialer Isolation auf die Gesundheit wurden jedoch Diskussionen um die ethische Legitimation und Verhältnismäßigkeit dieser Maßnahmen geführt. Ziel: Der Beitrag soll darstellen, in welcher Form in pflegewissenschaftlichen Publikationen der Diskurs zu COVID-19-Schutzmaßnahmen stattgefunden hat und welche inhaltlichen Aspekte dabei thematisiert wurden. Methoden: Es wurde ein Mapping Review in PubMed durchgeführt. Alle Beitragsformen wissenschaftlicher Publikationen zur pflegerischen Versorgung älterer Menschen wurden berücksichtigt. Die Ergebnissynthese erfolgte in Form einer quantitativen Inhaltsanalyse zentraler inhaltlicher Aspekte. Ergebnisse: Insgesamt zeigen die 38 in die Synthese aufgenommenen Beiträge, dass sich nur ein kleiner Teil der wissenschaftlichen Publikationen zur COVID-19-Pandemie mit Personen in der stationären Langzeitversorgung auseinandersetzt. Zwar werden in der Hälfte der Beiträge kritische Aspekte der Isolation bedingt durch die Schutzmaßnahmen vor der COVID-19-Pandemie benannt, jedoch werden nur selten konkrete Maßnahmen benannt, um gegen die entstehenden negativen Effekte durch die Isolation anzugehen. Schlussfolgerungen: Es bedarf weiterer Aktivitäten in Forschung und Pflegepraxis, um den Bedürfnissen und Bedarfen der Pflegebedürftigen gerecht zu werden und auch in einer Sondersituation wie der COVID-19-Pandemie Eigenverantwortung und Selbstbestimmtheit zu ermöglichen.

Virtual screening is a computational technique widely used for identifying small molecules which are most likely to bind to a protein target. In the present work, a molecular docking study is carried out to propose potential candidates for preventing the RBD/ACE2 attachment. These candidates are sixteen different flavonoids present in the peppermint leaf. Results showed that Luteolin 7-O-neohesperidoside is the peppermint flavonoid with a higher binding affinity regarding the RBD/ACE2 complex (about −9.18 Kcal/mol). On the other hand, Sakuranetin presented the lowest affinity (about −6.38 Kcal/mol). Binding affinities of the other peppermint flavonoids ranged from −6.44 Kcal/mol up to −9.05 Kcal/mol. The binding site surface analysis showed pocket-like regions on the RBD/ACE2 complex that yield several interactions (mostly hydrogen bonds) between the flavonoid and the amino acid residues of the proteins. This study can open channels for the understanding of the roles of flavonoids against COVID-19 infection.
