Core cross-linked polyphosphoester micelles with folate-targeted and acid-cleavable features for pH-triggered drug delivery

Abstract: Novel folate-conjugated acid-cleavable core cross-linked polyphosphoester micelles have been prepared and used for pH and enzyme-triggered delivery of doxorubicin.

“…This can be achieved either through non‐covalent loading in nanostructures with cleavable cross‐links or alternatively via macromolecular prodrugs, whereby the prodrug is covalently bound to the polymer. For example using acid‐cleavable acetal moieties as cross‐linkers for polyphosphoesters has been used to prepare drug‐loaded core‐cross‐linked micelles to prepare promising materials for targeted cancer chemotherapy.…”

“…This could be because of the longer time required for DOX to be released from micelles to tumor cells via an endocytosis mechanism, while free DOX entered into the cells by a passive diffusion process. 42 micelles could quickly release the encapsulated DOX upon redox stimuli.…”
