Cord Blood Mesenchymal Stromal Cell-Conditioned Medium Protects Endothelial Cells via STAT3 Signaling

Abstract: Background/Aims: Cell-based therapies may be useful for treating ischemic diseases, but the underlying mechanisms are incompletely understood. We investigated the impact of cord blood mesenchymal stromal cell (CBMSC)- or fibroblast (FB)-secreted factors on starved endothelial cells and determined the relevant intracellular signaling pathways. Methods: HUVECs were subjected to glucose/serum deprivation (GSD) in hypoxia or normoxia, in presence of CBMSC- or FB-conditioned medium (CM). Viability and proliferation… Show more

“…This is in line with the role of STAT3 in other systems (e.g., Bader et al, 2014). We find that absence of STAT3 results in increased Schwann cell death in four different situations.…”

“…Various different cell types are currently being investigated as potential cell therapy to improve neurological function after spinal cord injury or TBI including Schwann cells, fibroblasts and mesenchymal stem cells [30][31][32]. Effects of BMSCs on improving neurological functions have been well studied in recent years and studies are ongoing into methods of increasing their viability after transplant [33,34].…”

“…Therefore, the role of cytokines secreted by MSCs would assume importance in the cryoprotection of CD34 + cells. MSCs are known to secrete a variety of pro-survival factors in cardiovascular and neurodegenerative diseases, reducing apoptosis, inflammation and oxidative stress in these conditions [2427]. Thus, we sought to investigate if MSCs-CM would similarly exhibit a cryo-protective effect on the frozen HSCs.…”
