Copy number variant hotspots in Han Taiwanese population induced pluripotent stem cell lines - lessons from establishing the Taiwan human disease iPSC Consortium Bank

Abstract: Background: The Taiwan Human Disease iPSC Service Consortium was established to accelerate Taiwan's growing stem cell research initiatives and provide a platform for researchers interested in utilizing induced pluripotent stem cell (iPSC) technology. The consortium has generated and characterized 83 iPSC lines: 11 normal and 72 disease iPSC lines covering 21 different diseases, several of which are of high incidence in Taiwan. Whether there are any reprogramming-induced recurrent copy number variant (CNV) hots… Show more

“…With the improved reprogramming methods, various human cell types have been successfully applied to generate iPSCs. These include fibroblasts, peripheral blood mononuclear cells (PBMCs), T cells, B cells, hepatocytes, cord blood, dental pulp, skin, cardiomyocytes, and other somatic cells [ 18 , 20 ]. Theoretically, all cells in the human body have the potential to be reprogrammed to iPSCs.…”

“…Theoretically, all cells in the human body have the potential to be reprogrammed to iPSCs. With the advantages of being least invasive and easy to assess, PBMCs have become the most common cell source for human iPSC generation in biobanks [ 18 , 19 , 20 , 21 , 22 , 23 , 24 ].…”

“…The unprecedented story of anti-CD3 Muromonab OKT3 clone [ 5 – 8 ] for immune modulation upon transplant and its approval by the Food and Drug Administration (FDA) has paved the way to success for several antibody-based therapeutics. To that extent, antibodies and antibody-based therapeutics have become one of the fastest-growing treatment modalities within the modern drug arsenal, with a projected revenue of 300B USD by 2025 [ 9 ]. As of March 2020, the FDA has approved over 90 antibodies, and several of them (> 10) are under review for a variety of disease indications.…”
