Copper-Specific Chelators as Synergists to Herbicides:  1. Amphiphilic Dithiocarbamates, Synthesis, Transport through Lipid Bilayers, and Inhibition of Cu/Zn Superoxide Dismutase Activity

Abstract: Controlled inhibition of the enzyme Cu/Zn superoxide dismutase (SOD), one of the most important enzymes of the antioxidant defense system of aerobic organisms, is the subject of this paper. It is desirable in several noted medical and agricultural applications. This paper describes the synthesis of a very limited "sublibrary" of bifunctional metal chelators. The first function is the metal chelator DTC (sodium diethyldithiocarbamate (Et2DTC)), which has the sole chemical function of specific metal binding. An … Show more

“…Dithiocarbamates form particularly stable complexes with Cu 2þ . They are able to abstract the Cu 2þ cofactor from enzymes, leading to inhibition of enzymatic activity as was shown for the superoxide dismutase [226]. The dithiocarbamate-Cu complexes are membrane-permeable without causing leakages of the lipid bilayer, as was shown by stability experiments with liposomes [226].…”

“…It can be expected, however, that dissociated dodtc is distributed to the aqueous phase at higher pH values. Moreover, amphiphilic dithiocarbamates with longer alkyl chains have relatively lower critical micelle concentrations (cmc) in the range of 10 −6 −10 −7 M [9]. Indeed, %E values were improved when lower dodtc concentrations were employed (Fig.…”

“…The mechanism of action remains unclear but it is believed that interaction with oxyhemoglobin results in the formation of reactive radicals which spontaneously react with thiols resulting in intracellular glutathione depletion and other intracellular enzyme deactivation. Copper complexes of amino acids show SOD-like activity with those having longchain oligoether substituents being especially active [80] probably relating to their enhanced hydrophobicity. Copper(II) bis(dithiocarbamate) complexes find applications in radiopharmaceuticals.…”
