Copper‐Mediated Cascade Trifunctionalization of N‐Propargylamides

Abstract: The trifunctionalization of alkynes accompanied by intramolecular migration is one of the most powerful tools for rapid enhancing of molecular complexity in synthetic chemistry. Despite significant achievements have been made in the presence of noble metals (such as Au-, Rh-, Pd-, etc.), employment of copper is still underrepresented. Furthermore, 1,1-diiodoalkenes is the core structure of varieties natural products and pharmaceuticals, as well as valuable intermediates for futher manipulations. Herein we repo… Show more

“…Recently, increasing attention has been paid to tandem radical cyclization, which has emerged a major topic in sustainable chemistry. These reactions, which have been demonstrated as one of the most efficient routes to prepare isoquinoline and oxindole derivatives, can be initiated by radical addition to C–C double bonds and terminated with intramolecular radical cyclization of the aryl substituent.…”

“…In contrast, NO 2 -involved trifunctionalizations of alkenes have not been reported despite their potential advantage in the efficient and dense functionalization of alkenes in a one-pot manner (Scheme 1a). 8,9 As such, the development of novel reactions involving NO 2 radical-initiated trifunctionalizations of alkenes is highly desirable.…”

“…In particular, trifunctionalization of alkynes is an efficient procedure to achieve complex and diverse compounds by constructing three functional groups concurrently in one step . However, trifunctionalization of a terminal alkyne is challenging, and only a few papers on metal-catalyzed have been reported . Also, trifluoromethoxylation of alkynes is extremely rare.…”
