Coordinated transcriptional regulation of the carotenoid biosynthesis contributes to fruit lycopene content in high-lycopene tomato genotypes

Abstract: Lycopene content in tomato fruit is largely under genetic control and varies greatly among genotypes. Continued improvement of lycopene content in elite varieties with conventional breeding has become challenging, in part because little is known about the underlying molecular mechanisms in high-lycopene tomatoes (HLYs). We collected 42 HLYs with different genetic backgrounds worldwide. High-performance liquid chromatography (HPLC) analysis revealed lycopene contents differed among the positive control wild tom… Show more

“…Carotenoids have various health benefits, including antioxidant activity (Iannone et al., 1998), preventive effects on cardiovascular disease (Wang, Chung, et al., 2014), anti‐cancer effects (Kucuk et al., 2010), and anti‐aging effects (Kusmita et al., 2021). Lycopene is one of the carotenoids with relatively high antioxidant activity (Duduit et al., 2022). There are not many plants that can accumulate high levels of lycopene, which indicates its lycopene accumulation mechanism is extraordinary.…”

“…qPCR was performed in a CFX96 Touch Real-Time PCR detection system (Bio-Rad Laboratories; Hercules, CA, USA) using the FastStart Universal SYBR Green Master (Roche Diagnostics Corporation; Indianapolis, IN, USA) as described in Zhao et al . [ 77 ] and Duduit et al [ 92 ]. The optimal annealing temperature, primer concentration, and appropriate cDNA concentrations were determined to get the lowest Cycle threshold (Ct) value as the optimal conditions prior to the qPCR.…”
