Coordinated Expression of Astaxanthin Biosynthesis Genes for Improved Astaxanthin Production in Escherichia coli

Abstract: Astaxanthin has great potential commercial value in the feed, cosmetics, and nutraceutical industries due to its strong antioxidant capacity. In this study, the Escherichia coli strain CAR026 with completely balanced metabolic flow was selected as the starting strain for the production of astaxanthin. The expression of β-carotene ketolase (CrtW) and β-carotene hydroxylase (CrtZ), which catalyze the conversion of β-carotene to astaxanthin, was coordinated, and a bottleneck was eliminated by increasing the copy … Show more

“…The astaxanthin titer obtained by E. coli AST-4AS is lower than the highest value (1.18 g/L) ever reported [16]. The highest astaxanthin titer was obtained by high cell-density culture with OD600 of 469 [16]. However, the astaxanthin content (9.21 ± 0.12 mg/g DCW) obtained by E. coli AST-4AS is higher than that (7.8 mg/g DCW) reported in this literature [16].…”

“…E. coli AST-4AS produced astaxanthin of 311.20 ± 0.36 mg/L and 9.21 ± 0.12 mg/g DCW with a productivity of 5.55 mg/L/h, which was a 53.7% increase in the astaxanthin titer compared to E. coli AST-4. “…In such a pathway engineering research of these carotenoids, bacterial carotenoid biosynthesis ( crt ) genes have been utilized for their functional expression, e.g. crtE, crtB, crtI, crtY, crtZ and crtW genes have been used for astaxanthin production ( 15 , 19 , 20 ). Alternatively, few reports have described lutein biosynthesis in the E. coli host by the functional expression of carotenogenic genes that were derived from a plant (liverwort) (genes of lutein biosynthesis from lycopene) and bacteria (the crtE, crtB and crtI genes for lycopene biosynthesis) ( 21 , 22 ).…”
