Cooperative interactions of LFA-1 and Mac-1 with intercellular adhesion molecule-1 in facilitating adherence and transendothelial migration of human neutrophils in vitro.

Abstract: The adherence of human neutrophils to human umbilical vein endothelial cells (HUVEC) is partially dependent on the CD11/CD18 family of glycoproteins on the neutrophil and ICAM-1 on the HUVEC. The CD18 heterodimer involved in this adherence was evaluated in vitro using subunit-specific monoclonal antibodies (MAbs). The adherence of unstimulated neutrophils to IL-i-stimulated HUVEC was significantly inhibited by anti-CD11a but not CDlb MAbs, while the adherence of fMLP-stimulated neutrophils was significantly in… Show more

“…LFA-1, Mac-1 and p150,95 have different and yet overlapping roles in adhesion, in part due to their characteristics of expression on leukocytes. Stationary neutrophils adherent to the luminal endothelial surface frequently change shape and assume the characteristic bipolar configuration of motile cells [191,192]. This event may result from interaction with surface-bound chemokines.…”

“…20,55 Briefly, lung cryosections were stained with rat-anti-mouse CD18 antibody (1:100; BioLegend) and goat-anti-rat IgG conjugated with Alexa Fluor 488 (1:300; Invitrogen, Carlsbad, CA) for identification of neutrophils or monocytes in the lung tissue. Type I collagen, which is commonly found in lung and vascular tissue and is known to increase during acute injury and repair, 56,57 was identified using rabbit-antimouse collagen I antibody (1:100; Abcam) and goat-antirabbit IgG conjugated with Alexa Fluor 594 (1:300; Invitrogen).…”

“…Antibodies to ICAM-1 inhibit leukocyte adhesion to endothelial cells, granulocyte migration through endothelium, and mitogen and Ag-induced lymphocyte proliferation (Smith et al, 1988(Smith et al, , 1989. Crystal structures have been determined for domains 1-2 (D1-D2) and 3-5 (D3-D5) of ICAM-1 (Casasnovas et al, 1998;Yang et al, 2004), respectively, and the binding mechanism of ICAM-1 to its receptor, LFA-1, has been characterized in great detail (Jun et al, 2001a,b;Shimaoka et al, 2003).…”
