Convulsant effects of some xanthine derivatives in genetically epilepsy-prone rats

Abstract: The behavioural and electrocorticographic (ECoG) convulsant effects of several xanthine derivatives injected intraperitoneally (i.p.) were studied in genetically-epilepsy prone rats. The aim of the study was to evaluate the relationship among convulsant potency, molecular structure and lipophilicity of some xanthines. Animals were injected i.p. with various doses (250-1000 micromol/kg) and a different convulsant potency was observed among the various xanthines tested. IBMX (3-isobutyl-1-methylxanthine), theoph… Show more

“…The oral subchronic treatment with APs for 5 days in SD rats did not induce the appearance of audiogenic seizures following auditory stimulation in comparison with the SD control group, as previously described (data not shown) [26,51,52]. In GEPR-0s, haloperidol, quetiapine, and aripiprazole did not induce any change compared with the control GEPR-0 group at all doses used, whereas clozapine, risperidone, and olanzapine were able to induce the appearance of clonic phase of the audiogenic seizures (score 2-3) at the highest doses used (Fig.…”

“…Genetically epilepsy-prone rats (GEPRs) represent an established animal model to study the pathophysiology of seizures and to screen potential new antiepileptic drugs [24][25][26][27][28][29][30][31]. In this strain, epilepsy is genetically determined, even though the exact mechanisms underlying the development of the disorder remain incompletely understood [24,27].…”

“…Methylxanthines are used clinically as a diuretic, circulatory and respiratory stimulant, and treatment for headaches. The convulsive properties of methylxanthines have been well documented (14,15,29). Intoxication with caffeine and theophylline can lead to seizures at plasma concentrations Ͼ 100 M (30).…”

“…sistemici su altri organi ed apparati (8)(9)(10)(11)(12)(13)(14). Questa selettività d'azione della doxofillina sarebbe dovuta alla scarsa affinità con i recettori A1 e A2 dell'adenosina ed all'assenza di interferenze con i canali del calcio e con i relativi flussi ionici (8,13,14).…”
