Conversion of mouse fibroblasts to sphere cells induced by AlbuMAXI-containing medium

Abstract: The reprogramming of fibroblasts to pluripotent stem cells and the direct conversion of fibroblasts to functional neurons has been successfully manipulated by ectopic expression of defined factors. We demonstrate that mouse fibroblasts can be converted into sphere cells by detaching the fibroblast cells by protease and then using the AlbuMAX I-containing culture medium without genetic alteration. AlbuMAX I is a lipid-rich albumin. Albumin-associated lipids arachidonic acid (AA) and pluronic F-68 were responsib… Show more

“…AlbuMAX not only improved the viability and hatching ability of porcine blastocysts produced in vitro [ 14 ], but also regulated human embryonic stem cells (ESCs) self-renewal [ 15 ]. Sphere cells that have pluripotency could be induced from mouse fibroblasts in the culture conditions including AlbuMAX [ 16 ].…”

“…Mouse fibroblast cells were transdifferentiated into an intermediate state characterized by spherical cell aggregates, somewhere between mouse embryonic stem cells and mouse fibroblasts, using specialized medium containing lipid-rich albumin. Such spheres were able to differentiate into ectodermal germ layers following retinoic acid treatment[20]. microRNAs, such as miR9* and miR-124, participated in the transdifferentiation of fibroblasts into neuronal cells in the presence of other neurogenic transcription factors[2122].…”

“…During the same stage of reprogramming, NCs showed increased sterol biosynthesis (Fig ). In a previous study, murine embryonic fibroblasts could be transformed into sphere cells by suspension culture in lipid‐rich medium (Rajanahalli et al , ). Therefore, we theorized that the period prior to the MET process may represent a critical period for the removal of cell identity and that changing patterns of gene expression may provide insights into trans‐differentiation processes of different somatic cells.…”
