Controlled-release formulation of antihistamine based on cetirizine zinc-layered hydroxide nanocomposites and its effect on histamine release from basophilic leukemia (RBL-2H3) cells

Abstract: Abstract:A controlled-release formulation of an antihistamine, cetirizine, was synthesized using zinc-layered hydroxide as the host and cetirizine as the guest. The resulting well-ordered nanolayered structure, a cetirizine nanocomposite "CETN," had a basal spacing of 33.9 Å, averaged from six harmonics observed from X-ray diffraction. The guest, cetirizine, was arranged in a horizontal bilayer between the zinc-layered hydroxide (ZLH) inorganic interlayers. Fourier transform infrared spectroscopy studies indic… Show more

“…This result is similar to the kinetic study for the release of chlorogenic acid from ZLH layers, 13 camptothecin from Mg/Al-LDH, 46 and cetirizine from ZLH. 48 The correlation coefficient and release rate constant at pH 4.8 ( Figure 10E) are 0.9966 and 5.86×10…”

“…The intercalation of hippuric acid into ZLH has resulted in the less toxic of the compound compared to the pure hippuric acid itself [15]. It was also reported that the controlled release formulation of an antihistamine was obtained by forming the cetirizine-ZLH nanocomposite [16]. …”

“…The hippuric acid nanocomposite (HAN) was synthesized by the direct method using ZnO as the starting material as reported previously,9,13,14 with minor modifications. The hippuric acid solution (0.01 Mole) was prepared using 1.8 g of hippuric acid in 20 mL of DMSO, and was adjusted to 50 mL by the addition of deionized water.…”
