Control strategy of pH, dissolved oxygen concentration and stirring speed for enhancing β‐poly (malic acid) production by Aureobasidium pullulans ipe‐1

Abstract: BACKGROUND: β‐poly(malic acid) (PMLA) can be used as a pro‐drug or for a drug‐delivery system. Effects of pH, dissolved oxygen concentration (DO) and stirring speed were investigated to improve PMLA production by A. pullulans ipe‐1. RESULTS: The strain produced a high PMLA concentration when pH and DO remained at about 6.0 and above 70%, respectively, and the yeast‐like cells were the main PMLA producers. To further promote PMLA production, the cultivation could be divided into three phases. In phase I, cell g… Show more

“…However, L‐malate was widely verified and accepted as the precursor for PMLA biosynthesis . Meanwhile, a high DO level was beneficial for PMLA production . Therefore, the key enzymes activities involved in the precursor production for PMLA biosynthesis was studied under a high DO level (70% saturation) and a low DO level (10% saturation).…”

“…Meanwhile, Cao et al. found that a high dissolved oxygen (DO) level was beneficial for PMLA production. Moreover, other products biosynthesized by A. pullulans , such as pullulan and biosurfactant , were all affected by DO level.…”

“…In the 10-l benchtop fermentor, a high PMA concentration (57.2 g/l) produced by A. pullulans ZD-3 d was achieved within 160 h when the fermentation medium contained 120.0 g/l of glucose and 30.0 g/l of CaCO3 (Zhang et al, 2011). A. pullulans ipe-1 could produce up to 63.2 g/l of PMA during the repeatedbatch cultivation (Cao et al, 2013). A. pullulans strain ZX-10 produced PMA 50.0 g/l during batch fermentation in free-cell fermentation in a stirred-tank bioreactor (Zou et al, 2013).…”

“…That means the demand for oxygen in microorganisms varies among different fermentation processes. To ensure the appropriate oxygen supplement, DO is commonly controlled at a suitable level (Cao et al 2013). Take submerged fermentation of Saccharopolyspora spinosa , for example, DO is kept at 40–50% during 0–96 h for cell growth and at 25–35% during 96–240 h for spinosad formation (Bai et al 2015).…”
