Control of myeloid differentiation and survival by Stats

Abstract: Hematopoiesis involves a complex array of growth factors that regulate the survival and proliferation of immature progenitors, in¯uence di erentiation commitment, and modulate end-stage cell functions. This minireview is focused on the role of Stat activation in the development of myeloid cells in response to hematopoietic cytokines. Much of the evidence implicating Stats in these cellular processes comes from studies of mutant cytokine receptors selectively uncoupled from Stat activation, dominant-inhibitory … Show more

“…Although our findings do not distinguish between these possibilities, they further highlight the role of STAT3 in antiapoptotic signalling. Hirano et al, 2000;Smithgall et al, 2000). In addition, STAT proteins are constitutively active in many leukaemias (reviewed in Lin et al, 2000).…”

“…STAT3 is a pleiotropic transcription factor that influences proliferation, differentiation and survival of multiple cell types. It can induce proliferation by promoting expression of its transcriptional targets Cyclin D or c-Myc, but paradoxically, STAT3 activation is cytostatic in other cell types via the induction of cell cycle inhibitors such as p21 CIP , p27 KIP or p14 Arf (reviewed in Bowman et al 31 and Smithgall et al 32 ). This dual mode of action is perhaps best illustrated by the opposing effects of activated STAT3 on tumorigenesis.…”

“…The roles for STAT3 in G-CSF-induced granulocyte differentiation and in CNTF-and OSM-induced astrocyte differentiation in vitro have also been reported. [30][31][32] Recently, constitutive activated STAT3 in myeloma was suggested to confer resistance to apoptosis. 14 From this study and our previous studies, 9 we show that IL-6 induces a proliferative response in the malignant plasma cells by inducing expression of cyclins, repressing cdk inhibitors, and up-regulating c-myc expression 33 ( Figure 5).…”
