Control of Developmental Regulators by Polycomb in Human Embryonic Stem Cells

Abstract: Polycomb group proteins are essential for early development in metazoans, but their contributions to human development are not well understood. We have mapped the Polycomb Repressive Complex 2 (PRC2) subunit SUZ12 across the entire nonrepeat portion of the genome in human embryonic stem (ES) cells. We found that SUZ12 is distributed across large portions of over two hundred genes encoding key developmental regulators. These genes are occupied by nucleosomes trimethylated at histone H3K27, are transcriptionally… Show more

“…By contrast, Meis1 and Fli1, two genes that are normally repressed by PRC2 (Lee et al, 2006), were actively transcribed in all investigated t(4;11) biopsy probes. This may indicate that specific Nanog/Oct4 target genes may be deregulated in the presence of the t(4;11) fusion proteins.…”

“…[19][20][21] Boyer et al reported the genes occupied by PRC1 (Phc1 and Rnf2) and PRC2 (Suz12 and Eed) in murine ESCs. 19 To explore a novel target of Bmi1 in hepatic stem/progenitor cells, we compared the list of down-regulated genes with the ChIP-on-chip data documented by Boyer et al 19 As a result, five genes namely, Sox17, Irx5, Gjb2, Shox2, and Bhmt2 in the present study appeared to be regulated by both PRC1 and/or PRC2 in ESCs (Fig.…”

“…But our ChIP assay showed that siSnail, but not siYY1, dissociated the recruitment of EZH2, HDAC1/2 from this region. Several groups had reported that the PcG proteins and H3K27me3 were tend to be broadly distributed across many kilobases of target gene locus Lee et al, 2006;Schwartz et al, 2006;Simon and Kingston, 2009). An alternative interpretation is that the large distribution of PcG complex is account for the looping action mechanism, leading PcG complex to be crosslinked to more distant nucleosomes (Schwartz and Pirrotta, 2007;Tiwari et al, 2008).…”
