Control of Cell Size by c-di-GMP Requires a Two-Component Signaling System in the Cyanobacterium Anabaena sp. Strain PCC 7120

Abstract: Cyanobacteria are considered the first organisms to produce oxygen on Earth, and their activities shaped the evolution of our ecosystems. Cell size is an important trait fixed early in evolution, with the diversification of micro- and macrocyanobacterial species during the Great Oxidation Event.

“…Our results also support the recent research that "cheaters" of QS were capable of active communication by recruiting organisms from other domains, forming a pair in which one organism carries the receptive gene and another the synthetic gene [16]. Additional QS communications in bacteria are intricately connected to AOA, such as the AI-2 system and cyclic-di-GMP system; the latter has been reported as a signal modulating the cell size of cyanobacteria [71,72]. The interplays observed between AOA metabolic pathways and bacterial quorum sensing underscores the essential role of communication in metabolic exchanges in the marine water column, which could enhance the growth efficiency of AOA and surrounding heterotrophic communities.…”

“…shown in Figure 3a, whatever the OD of the culture, cell elongation was observed under diazotrophic conditions, including the culture maintained at an OD 750 value of 1.0. With the same cell concentration (OD 750 = 1.0), cells of the WT became even smaller in length under diazotrophic conditions, as previously described (Neunuebel & Golden, 2008;Sun et al, 2023). Consistently, filaments of the mutant appeared yellowish in colour in comparison to those of the WT (Figure 3b), and further incubation under such conditions led to cell lysis.…”

“…This multi-domain protein (CdgSH) harbours a GAF domain that does not contain the amino acids important for chromophore binding in a typical photosensory GAF domain. Further, it has been shown that a phosphodiesterase (CdgS), which is part of a two-component system including the multi-domain histidine kinase CdgK, controls the cell size of this cyanobacterium (Sun et al 2023 ). In both systems, the signal for the activation of c-di-GMP-related enzymes is unknown.…”
