Control of Allergen-Induced Inflammation and Hyperresponsiveness by the Metalloproteinase ADAMTS-12

Abstract: A disintegrin and metalloproteinase with thrombospondin motifs (ADAMTS) constitute a family of endopeptidases related to matrix metalloproteinases. These proteinases have been largely implicated in tissue remodeling associated with pathological processes. Among them, ADAMTS12 was identified as an asthma-associated gene in a human genome screening program. However, its functional implication in asthma is not yet documented. The present study aims at investigating potential ADAMTS-12 functions in experimental mo… Show more

“…In addition, the antiangiogenesis properties of ADAMTS-12 did not rely on the catalytic domain, but, more likely, on its TSP-1 domains [30]. In contrast, the catalytic activity was implicated in the protective effect against allergen-induced inflammation [28]. …”

“…There has been increasing evidence pertaining to the pathological relevance of this enzyme. For instance, ADAMTS-12 has been proven to be associated with the pathogenesis of arthritis [816], intervertebral disc degeneration [25, 26], inflammation [27, 28], and invasion and metastasis of tumor [2931]. Although ADAMTS-7 and -12 share similar structure and domain organization, ADAMTS-7 and -12 may have different pathological effects or be responsible for different phases in a disease [26, 32].…”

“…For antigen retrieval, the slides were boiled in 10 m M citrate buffer (pH 6.0) for 10 min in a microwave [14]. Digital images were acquired using an Olympus BX50 microscope (Olympus Europa GmbH, Hamburg, Germany) equipped with a Leica 320 digital camera (Leica Microsystems, Wetzlar, Germany).…”

“…ADAMTS12 gene is found in skeletal muscles, cartilage, tendons and fetal lung. [7][8][9] ADAMTS12 was shown to be associated with arthritis, 10 intervertebral disc degeneration, 11,12 inflammation, 13,14 tumor invasion, and metastasis. [15][16][17] Also, high levels of ADAMTS12 messenger ribonucleic acid (mRNA) were detected in osteoarthritis and rheumatoid arthritis.…”
