Control and maintenance of mammalian cell size

Abstract: BackgroundConlon and Raff propose that mammalian cells grow linearly during the division cycle. According to Conlon and Raff, cells growing linearly do not need a size checkpoint to maintain a constant distribution of cell sizes. If there is no cell-size-control system, then exponential growth is not allowed, as exponential growth, according to Conlon and Raff, would require a cell-size-control system.DiscussionA reexamination of the model and experiments of Conlon and Raff indicates that exponential growth is… Show more

“…These mechanisms are distinguishable through analysis of DNA content and cell size (reviewed in (Cooper, 2004)). To determine which of these mechanisms contribute to the proliferative arrest of TRPM7-deficient cells cultured in regular media, we analyzed the DNA content (via DAPI staining) and cell size (via forward scatter) of TRPM7-deficient cells upon transition from proliferation in growth supporting Mg 2+ -supplemented media to regular media (Figure 1A).…”

“…Our model assumed the EGF signaling pathway in a mammalian cell in which the volume is typically > 10 −12 L, which is much larger than yeast or bacteria (10 −16 –10 −14 L) [45,46]. Therefore, a mammalian cell has a huge number of molecules even if the same concentrations of proteins are present in yeast and bacterial cells.…”

“…A logarithmic fit was used to determine the equation that permits calculating the concentration in the whole cell lysates. Finally, an estimation of cell volume70 was used to determine the concentration of DWNTs per cell.…”
