We previously reported the inhibitory potentials of momilactones A (MA) and B (MB) against key enzymes related to type 2 diabetes and obesity. In this study, antioxidant and anti-skin-aging activities of MA and MB were investigated and compared with tricin, a well-known antioxidant and antiaging flavonoid in rice. MA, MB, and tricin were purified from rice husk by column chromatography and their biological activities were subsequently assayed by in vitro trials. The contents of MA, MB, and tricin of different commercial rice cultivars in Japan were quantified and confirmed by ultra-performance liquid chromatography-electrospray ionization-mass spectrometry (UPLC-ESI-MS) and high-performance liquid chromatography (HPLC) analyses. The antioxidant assays revealed a synergistic activity of the mixture MA and MB (MAB, 1:1, v/v). In addition, in 2,2'-azino-bis (ABTS) assay, IC 50 values of MAB (0.3 mg/mL) and tricin (0.3 mg/mL) was 4-fold and 9-fold greater than that of individual MB (1.3 mg/mL) or MA (2.8 mg/mL), respectively. The in vitro enzymatic assays on pancreatic elastase and tyrosinase indicated that MA and MB were potential to relief skin wrinkles and freckles. In detail, MA exerted higher inhibition on both enzymatic activities (30.9 and 37.6% for elastase and tyrosinase inhibition, respectively) than MB (18.5 and 12.6%) and MAB (32.0 and 19.7%) at a concentration of 2.0 mg/mL. Notably, MA and the mixture MAB exhibited stronger inhibitions on elastase and tyrosinase in comparison with tricin and vanillin. MA, MB, and tricin in rice are potential to develop cosmetics as well as supplements for skin aging treatments.

Astragalus kurdicus Boiss. roots are used in folk medicine for antidiabetic purposes. Different metabolites of Astragalus plants have a notable potential in antidiabetic activity via differing mechanisms. Herewith this study designed to assess the antidiabetic activity of Astragalus kurdicus, utilizing a range of diabetes‐related in vitro methodologies and to investigate the chemical composition of the plant. According to the results of the activity tests, water extract (AKW) was the most active extract in PTP1B, DPP4, and α‐amylase inhibition tests (87.17%, 82.4%, 91.49% respectively at 1 mg/ml). Total extract, AKM (85.63%), showed the highest AGEs inhibition activity. To test possible improvement effects of the extracts on diabetes through gut‐microbiota, cell growth rates of three probiotic microorganisms were measured. AKM showed highest potential of prebiotic activity among tested extracts and caused higher biomass increase than standard prebiotics. Furthermore, flavonoid‐rich extract was found to be mostly responsible for the high antioxidant activity. The highest saponin and astragalosides content were seen in AKB extract in HPTLC analysis. Among the measured saponins, the abundance of Astragaloside IV (27.41 μg/mg in AKM) was the highest in all fractions. Thus, for the first time, the antidiabetic activity of A. kurdicus was evaluated from various perspectives.

Diabetes mellitus is the most serious metabolic disorder in the world. 1 A recent report from the World Health Organization (WHO) showed that more than 422 million people worldwide have diabetes, especially in low-and middle-income countries. Diabetes mellitus can be divided into two types: insulin-dependent (type 1 diabetes mellitus) and non-insulin-dependent (type 2 diabetes mellitus). The common patients with diabetes are type 2 patients. Type 2 diabetes mellitus is caused by high blood glucose level which is hydrolysed from starch. 2 α-Glucosidase and α-amylase are the group of starch hydrolytic enzymes. The polysaccharides hydrolyse to short oligosaccharides or disaccharides by α-amylase, and finally, the free glucose is released from disaccharides by α-glucosidase. 3 The inhibiting both α-glucosidase and α-amylase has reported by varied plant bioactive compounds. Nazir et al 4 report that the chloroform extract of Elaeagnus umbellata (Thunb.) has shown high inhibitory potential on both α-glucosidase and α-amylase. Capsaicin and dihydrocapsaicin Abstract Coloured rice is the source of bioactive compounds related to medicinal treatment.This study aimed to investigate α-glucosidase and α-amylase inhibitory activity and antioxidant properties of purified coloured rice extract (Thai fragrant black rice). The results reveal that seven purified fractions; rice fractions 1-7 (RF1-RF7) contained several phytochemical compounds especially flavonoids, which were presented in all purified fractions. The percentage inhibition on α-glucosidase and α-amylase of fractions RF1, RF2, RF4, RF5 and RF6 had higher potential than acarbose. RF4, RF5 and RF6 contained the highest inhibition potential, which further determined the IC 50 values, kinetic analysis and antioxidant properties. The IC 50 values of all three fractions indicated that the inhibition potential of purified fraction had higher potential than acarbose, and the highest inhibition potential for both enzymes was obtained from fraction RF5. The kinetic analysis of the three fractions indicated that they are against both enzymes' function in mixed-type inhibition. Moreover, all three fractions also showed good antioxidant activity, and the highest potential was obtained from fraction RF6. This study presents the inhibition efficiency of fragrant black rice extract on α-glucosidase and α-amylase enzymes. The inhibitory activity of rice extract on both enzymes had higher potent than commercial standard drug acarbose.Moreover, the rice extract also shown the high-efficiency antioxidant activity on DPPH and ABTS assay. We suggest the application of these results in the development of anti-diabetes drug for pharmaceutical industry.
