Contribution of dendritic cells to stimulation of the murine syngeneic mixed leukocyte reaction.

Abstract: We have studied the proliferative response of unprimed T cells to syngeneic dendritic cells (DC) (syngeneic mixed leukocyte reaction [SMLR]) in cultures of mouse spleen and lymph node. T cells purified by passage over nylon wool contain few DC and exhibits little proliferative activity during several days of culture. Addition of small numbers of purified syngeneic DC induces substantial, dose-dependent, T cell-proliferative responses that peak at day 4-5. B cells purified on anti-Ig-coated plates do not respon… Show more

“…SDC from CBA/H mice induced high levels of background T cell proliferation, in the absence of WNV antigen, at tbe two top S:R ratios used, in keeping with potent syngeneic mixed leucocyte reactions previously reported by Nussenzweig and Stcinman (34). In contrast, the responses induced by DC from C57BL/6 mice were comparable with those obtained using B|4 or Bo APC.…”

“…Dendritic cells, preferentially of a mature phenotype, are considered to be the prime initiators of such responses. 35,36 Alternatively, the responding T cells may recognize foreign antigens engulfed during ex vivo DC-generation. As no serum was added in our protocol, the candidate antigens would be derived from supplemented cytokines, like GM-CSF, 37 or proteins in the Cellgro DC-medium.…”

“…DCs have long been assumed to be the prime triggers of AMLR. 21 In our restimulation cultures, potential AMLR antigens are effectively presented by large numbers of mature DCs. Alternatively, the T-cell response to mockDCs could be elicited by foreign antigens engulfed during DC culture.…”
