Contraceptive Effect of Sperm-Agglutinating Monoclonal Antibodies in Rabbits1

Abstract: Immune infertility in humans correlates clinically with the presence of anti-sperm antibodies that trap (agglutinate) sperm in semen and cervical mucus. To test whether sperm-agglutinating antibodies can be effective contraceptive agents, several mouse anti-rabbit sperm (MARS) sperm-agglutinating monoclonal antibodies (mAbs) were developed that rapidly and completely agglutinate sperm: MARS-M3 (IgM), MARS-G16 (IgG3), and MARS-G17 (IgG3). Contraceptive efficacy of these mAbs was tested by mixing the mAb with 0.… Show more

“…Indeed, an anti-rabbit sperm mAb has been shown to provide contraceptive activity in a rabbit model. 32 A mAb (HC4) against a surface glycolipid present on human sperm has been produced in the ΔXF Nicotiana line and shown to co-agglutinate 100% of sperm and other seminal cells in less than thirty seconds at 100 ug/ml (Fig. 2).…”

“…This may closely mimic the normal function of antibodies in the mucosal immune system, e.g., antibodies in breast milk help protect the surfaces of the mouth, nose, eyes, and digestive tract of the baby. To date, the results of studies performed indicate that monoclonal antibodies delivered to the vagina may help prevent pregnancy as well as sexual transmission of genital herpes and HIV (161)(162)(163)(164).…”

“…Agglutination of rabbit sperm with unformulated IgM Ab has been shown to provide contraceptive activity in a rabbit model (Castle et al, 1997); this study mimics the agglutination mechanism that is associated with immune infertility in humans (WHO, 1992). A Nicotiana manufactured IgG 1 against a unique (found only in the human male reproductive tract) glycoform of CD52, i.e.…”
